Two parts are included in this thesis: Studies on in vitro and in vivo biotransformation of (-)shikonin and studies on the detection and determination of some lignans in the seeds of Arctium lappaol Linne.1.The metabolic biotransformation of shikonin was studied. First, the in vitro metabolism of shikonin was investigated, a HPLC method for the detection of the metabolites of shikonin was developed and used to optimize the metabolic system of shikonin and to study the in vivo biotransformation. The research was mainly concerned with the following subjects:(1)The in vitro incubation metabolic system catalyzed by rat liver microsomes was adopted to study the metabolism of shikonin. Gradient and Socratic RP-HPLC/DAD methods were developed to separate and detect the metabolites. The effect of several factors(such as composition of incubation system, administration mode, metabolic time and microsomes induced by PB or not) on the results of metabolism were investigated, and the metabolic system was optimized. The metabolic results of S9 and microsome incubation system were compared. The results showed that there were 7 metabolites of which the UV spectra are similar to that of shikonin, the contents of three of them with retention times of 3.55min,6.92min,7.99min respectively, are higher than the others.(2)A LC-ESP-MS method was developed and used to directly separate the in vitro incubation extracts of shikonin and the structures of metabolites were proposed. The column, stationary phase, composition and flow rate of the mobile phase were optimized to fit LC-MS.
