Background and objective Cervical squamous cell carcinoma (CSCC) is the most common malignant tumor for women . Moreover, the incidence of CSCC for young women is rising in recent years. But specific protein biomarkers were not detected using for auxiliary diagnosis , evaluating the effectiveness of therapy and prognosis of cervical cancer. The specific protein biomarkers associated with oncogenesis and progression, or with the treatment and prognosis of cervical carcinoma are screened by proteomic study . Methods The tissue lysate from six matched pairs of cancer /normal tissues of CSCC had been profiled and identified via 2-DE and 2-DE/Western blot analysis. Differentially expressed proteins were confirmed additionally by Western blot and immunohistochemistry staining. The expression, distribution and localization of the protein of CSCC were assayed. Results Analysis of 2-DE images from protein lysates of 6 pairs CSCC detected 386 protein /peptide spots in cancer tissues and 322 protein/peptide spots in matched normal tissues, and 45 protein/peptide spots detected, were different and matched with the reference gel. Of 45 protein/peptide spots, Grp94(glucose regulated protein,Grp94) is significantly a different protein with a good repeatability between tumor tissues and normal epithelial tissues. Grp 94 was up-regulated in CSCC by WB(western blot) assay (94.4%). Grp 94 was positively expressed in 93.9% of CSCC by imunohistochemistry staining, which is diffusely distributed in the cytosol. But it was negatively expressed in cervical intraepithelial neoplasias and normal squamous epithelial cells.
