Background: Cervical cancer(cervical cancer), also called cervical cancer, is located in the female genital tract malignant tumor incidence and mortality of(1). According to the pathological types can be divided into squamous cell carcinoma, adenocarcinoma and gland scale cancer, with squamous cell carcinoma, most accounts for 80% to 80%. The onset of cervical cancer type mainly with high-risk human papilloma virus(human papilloma virus comes, HPV) persistent infection, at the same time with more premature sexual activity, sexual life, delivery times, smoking, socioeconomic status, immune deficiency and other factors. Progress of cervical cancer is a multi-factor, many links, many kinds of protein molecule involved in multi-step process, it is a serious threat to the women’s health and quality of life, so the cervical cancer early detection, early diagnosis, early treatment is particularly important. 39 YKL- 40 also known as human cartilage protein, is a kind of mammalian enzyme sample protein shell material. YKL-40 since was first found in 1988, a growing number of experiments show that YKL-40 in a variety of solid tumor can be detected abnormal expression, and that its in the body tissues may be involved in tumor cell growth, differentiation, invasion and metastasis, tumor the reconstruction of the extracellular matrix and new angiogenesis, inflammatory reaction and other life activities. CD 105(Endoglin) is a kind of proteins related to the new vascular endothelial cell proliferation, and the formation of new blood vessels is the important mechanism of tumor development. Studies show that CD 105 in liver cancer(2), colorectal cancer(3), of breast cancer(4), ovarian cancer(5),cervical cancer(6), endometrial cancer(7) and so on the many kinds of cancer are abnormal expression in blood and tissue. Hint it is closely related to the occurrence and development of malignant tumor.Objective:The purpose of this paper is to detect YKL- 40, CD105 in normal healthy women, cervical intraepithelial neoplasia(CIN), the palace expression in cervical squamous cell carcinoma, and to explore the two in cervical squamous cell carcinoma of the interaction relations and its clinical significance.Methods: Respectively using enzyme-linked immunosorbent assay(ELISA double antibody clip art) detection of 52 cases of the two aspects of clinical and pathological diagnosis of cervical squamous cell carcinoma patients with stage I/II(line can be directly after the first diagnosis surgery), according to the results of preoperative staging and the postoperative pathological slow returns with and without lymph node metastasis and pathological differentiation level data statistics, including 40 cases in stage I, stage II 12 cases; Without lymphatic metastasis, 41 cases with lymph node metastasis in 11 cases; High- differentiation in 38 cases, 14 cases of low differentiation; 50 cases of patients with cervical intraepithelial neoplasia(CIN), including CINI class 20 cases, CINII level of 18 cases, 12 cases CINIII level; 30 cases of normal healthy women(control group) in serum of YKL- 40 and the expression of CD105. All the research object in the first option on the same day in the morning on an empty stomach condition extraction of venous blood 3 ml, including cervical cancer group, all patients underwent early in the morning on an empty stomach condition on the 7th of extraction of venous blood again 3 ml, are placed without anticoagulant vacuum in the bloodstream. Will collect all the blood in room temperature, solidification 55 minutes. After being serum levels of natural precipitation, in 4 ℃environment, at a speed of 2000 r/min, the centrifugal 10 minutes, drain upper serum, moved to the EP tube cleaning, completes the tag, at 80 below zero℃ saved for later use. All samples need to avoid repeated freezing and thawing, hemolysis. All data will get application SPSS19.0 package for statistical analysis.Results:1. The control group, cervical intraepithelial neoplasia and cervical cancer patients with preoperative serum YKL-40 and CD105 concentration mean concentration with the development of lesions showed a trend of increase, cervical cancer concentrations were significantly higher in patients with cervical intraepithelial neoplasia group and normal control group, the difference was statistically significant(P < 0.05); Cervical intraepithelial neoplasia between groups was no significant statistical difference(P > 0.05); Cervical intraepithelial neoplasia group compared with normal control group no significant statistical difference(P > 0.05). YKL-40 and CD105 in the normal control group, cervical intraepithelial neoplasia and cervical cancer group in the positive expression rate gradually increased, the expression rate of cervical cancer group was obviously higher than that of the rest of the two groups, the difference was statistically significant(P < 0.05) in cervical intraepithelial neoplasia group and normal control group no significant difference(P > 0.05).2. I/II cervical cancer group serum YKL-40 before and after operation, the expression of CD105 horizontal comparison, both the concentration of the postoperative mean differences were less than before, and the difference was statistically significant(P < 0.05).3. YKL-40 concentrations are associated with pathologic stage, lymph node metastasis, namely low differentiated expression in cervical squamous cell carcinoma patients with serum concentration higher than that of high differentiation and differentiation in patients, with lymph node metastasis is serum YKL-40 concentration higher than without lymph node metastasis, the difference was statistically significant(P < 0.05); Has nothing to do with FIGO stage(P > 0.05), there was no statistically significant difference. And the concentration of CD105 and FIGO staging, pathologic stage, with or without lymph node metastasis are related(P < 0.05).4. Serum YKL-40, CD105 was positively expressed in cervical squamous cell carcinoma patients serum in the correlation(r = 0.265).Conclusion:(1) YKL-40 and high expression of CD105 may in has played a certain role in the occurrence and development of cervical cancer.(2) YKL- 40 and CD105 may be involved in the transfer of cervical cancer, the infiltration process.(3) YKL-40, CD105 evaluation of cervical cancer surgery has certain reference value.(4) The YKL-40, CD105 both joint detection for early diagnosis of cervical cancer has a certain value.
