Objective: By analysis of various " perennial root" theories,with the aid of new understanding of modern medicine on "perennial root" concept and on the basis of clinical and experimental study, the new hypothesis of "spleen weakness is the perennial root of asthma " was explicitly proposed for the first time.By taking reinforcing earth to strengthen metal as the principle ,the decoction of strengthening the spleen and tonifying the lung was used; with modern molecular biological method ,the effects on the morphology of eosinophil apoptosis and Fas mRNA, Bcl-2 the mRNA expression in rats with spleen deficency asthma were observed. The effects of spleen deficiency on asthma and the effective mechanism of recinforcing earth to strengthen metal on asthma were explored.Methods: Under the guideline of the theory of traditional Chinese medicine(TCM), the rat models with spleen-deficiency in TCM and bronchial asthma in west medicine were set up. After the rat models were formed successfully, the decoction of reinforcing earth to strengthen metal was used for treatment. Apoptosis was detected by TUNEL,and Fas mRNA and the Bcl-2 mRNA expressions in the tissue of lung were detected by hybridization in situ and the concentration of IL-5 and GM-CSF in BALF were detected by enzyme linked immuoabsorbent assay (ELISA).The morphology of the lung tissue was observed by the light microscop.Results: Compared with control group, in both groups of the combination of disease with syndrome group and the asthma group, the levels of GM-CSF remarkably increased(P <0.01) and the levels of TGF-β remarkably decreased in BALF. Compared with control group, the count of EOS increased and EOS apoptosis rates decreased (P <0.01 or P<0.05).Compared with control group, in both groups of the combination of disease with syndrome group and the asthma group,the expressions of Fas mRNA remarkably decreased and the expressions of Bcl-2 mRNA remarkably increased (P <0.01). At the same time.compared with the corresponding asthma group,in both decoction groups(Al,Bl), Bcl-2mRNA expression was remarkably reduced (P <0.01), Fas mRNA expression obviously increased(P <0.01 or P < 0. 05). A positively significant correlation was found between the Fas mRNA expression and EOS apoptosis in each group (r=0.62, p<0.05); and a very significant negative correlation was found between the Bcl-2mRNA expression and EOS apoptosis (r= - 0.58, p<0.05) ; a positively significant correlation was found between the EOS apoptosis and the concentration of TGF-P in BALF in each group rats (r=0.51, p<0.05) .The results indicated a very significant negative correlation of the EOS apoptosis with the concentration of GM-CSF in BALF(r= - 0.65, p<0.01).
