Part ⅠComparison in biological characteristics of fibroblasts derivedfrom human oral normal mucosa and oral cancer stromaObjective To investigate the differences in cellular biology characteristics of fibroblasts derived from human oral normal mucosa and oral cancer stroma. Methods Invasive oral squamous carcinoma tissue and its corresponding resection margin were obtained from the same patient. After 3-8 passages, the oral carcinoma-associated fibroblasts (CAFs) and normal mucosa fibroblasts(NMFs) from tissue culture were identified under light microscope, transmission electronmicroscope and Vimentin, α -smooth muscle actin( α -SMA) and Cytokeratin proteinwere detected by immunocytochemistry SP method on glass coverslips with CAFs and NMFs grown. Oral CAFs and NMFs were compared of their proliferation characteristics, such as growth curve , doubling times and mitotic index curve to assay the vitality of the two kinds of cells. A growth-promoting effect was observed with serum-free conditioned medium from oral CAFs and NMFs. The expression of genes...
