Objective This experiment was designed to study the effect of dichlobenil and sodium deoxycholate on nasal mucosa including both respiratory and olfactory region ,and to construct the method for systematic evaluation of drug safety to nasal mucosa.Methods Use dichlobenil and sodium deoxycholate as test article, wistar rat and beagle dog as test system to study their effect to nasal mucosa such as respiratory region and olfactory region. 1. (a)the isolated nasal mucociliary cells from normal animals were put into sodium deoxycholate at concentration of 0.0001g/ml, 0.001g/ml, 0.01g/ml, and observed and video-recorded under microscope, and ciliary beat frequency was calculated, (b) Isolated nasal mucociliary cells from animals treated with sodium saline or 10 μ10.01g/mL sodium deoxycholate by intranasal dropping were put into sodium saline and observed and video-recorded under microscope, and ciliary beat frequency was calculated. 2. beagle dogs were treated by nasal spray of saline sodium or 100 μl 0.01g/mL sodium deoxycholate for 7 consecutive days once a day, Dog nasal respiratory mucosa were drawn and evaluated by biological light, scanning and transmission electron microscopy.
