Cholangiocarcinoma is a kind of cancinoma with poor prognosis because its lately symptom emergence, hardly early diagnosis, low radical cure rate and poor insensitivity chemotherapy drugs. Tumorigenesis and its development is not only the result of inordinate cell proliferation and abnormal differentiation but also the disbalance of apoptosis. So it a good tumor therapy strategy to interfere apoptosis of tumour cell. Paclitaxel has become a new important chemotherapy drug for its distinctive pharmacologic role and has been used generally in clinical treatment. But the mechanisms of its role are still unknown .They need further research. With the improvement of polyacrylamide gel electrophoresis and mass-spectrum technique, proteome analysis has become a important method for clinical disease research. To investigate Tumorigenesis mechanism of cholangiocarcinoma, illuminate new targets for chemohteray and provide new theory foundation for clinical medication, we chose apoptosis as our research directions and investigated the apoptosis mechanism of cholangiocarcinoma cell induced by paclitaxel. In this study, cholangiocarcinoma QBC939 cell lines were induced apoptosis by paclitaxel with applied cell culture, observed cell morphous, analyzed the cell cycle and proteome technique was employed to analysis the expression of protein spectra difference between cells treated with paclitaxel group and control.Chapter I Assay sensibility of cholangiocarcinoma QBC939 celllines to paclitaxel in vitroObjective: To investigate the sensibility of cholangiocarcinoma QBC939 cell lines to paclitaxel and find new chemotherapy drug for cholangiocarcinoma.
