Studies On The Chemical Constituents And Anticancer Activity Of Bauhinia Variegata L.

In the present dissertation, 21 species of plants with anticancer activity screened by MTT method were examined again using tsFT210 cells, a mouse temperature sensitive cdc2 mutant cell line. The result suggested that 10 species still showed various degrees of activity. Among them alcohol extract of the stems of "Yangzijing", showed not only good G₂/M phase inhibitory and apoptosis activities to tsFT210 cells but also good G₀/G₁ phase inhibitory and apoptosis activities to human myeloid leukemia K562 cells. "Yangzijing", Bauhunia variegta L., belonging to Leguminosae, has been used as a folk medicine in Guangxi, Yunnan province of China to cure indigestion, acute gastroenteritis, bronchitis, tracheitis, pneumonia, pulmonary tuberculosis, hepatitis, etc. It has also been used to cure cancer in some regions of Yunnan province. Up to now, few literatures on the chemical constituents of Bauhinia variegate L. have been reported and no bioactivity research has been seen yet.In order to elucidate the anticancer constituents and lay foundation for development and enlargement of the medicinal sources of Bauhinia variegate L., the chemical and biological investigation had been carried out by the numbers. The results are as follows:1. Guided by bioassay of cell cycle inhibitory and apoptosis inducing activities using tsFT210 and K562, the active parts and fractions had been determined and the isolating scheme for active components had been established. 22 chemical compounds had been isolated by means of silica gel, ODS and Sephadex LH-20 column chromatogram and identified on the basis of the physicochemical properties and spectra data as N-naphthyl-2-aniline (1), biocyclomahanimbine (2), mahanimbine (3), grinimbine (4), bauhinione (5), physcion (6), epigallocatechin (7), protocatechuic acid (8), protocatechuic acid ethyl ester (9), methyl gallate (10), ethyl gallate (11), p-hydroxybenzonic acid (12), friedelin (13), 24 (R) - 9,19 - cyclolanost - 3 - one - 24 , 25 - diol (14), 24 (S) - 9, 19 -cyclolanost - 3 - one - 24, 25 - diol (15), stigmast - 3 - one (16), stigmast - 4 - en - 3 - one (17), stigmast - 3β - ol - 6 - one 18), stigmast - 5 - en - 3β - ol - 7 - one (19), schizandriside...
