In this study,Uncaria rhynchophylloides How and Bauhinia touranensis Gagnep were taken as research objects,and systematic chemical composition analyses were carried out.Furthermore,we aimed to assess potential in vivo and in vitro antihypertensive activities of U.thynchophylloides.61 compounds were isolated from Bauhinia touranensis and Uncaria rhynchophylloides How,with 4 new compounds,including alkaloids,flavonoids,lactones,iridoids,triterpenes,phenylpropanoids and the other phenolic constituents.The pharmacological results suggested there might be a superior antihypertensive activity of U.rhynchophylloides,for which contains plenty of iridoids and phenolic constituents.The first chapter introduces the research on the chemical composition of U.rhynchophylloides,and 25 compounds were isolated and identified from the 95%ethanol extract of the stems and branches of U.rhynchophylloides,including a new compound(Howlumine).Two 1,4-diazaindan-type alkaloids(1-2),three iridoid glycosides(3-5),four megastigmane glycosides(6-9),two phenylpropanoids(10-11),two benzoic acid derivatives(12-13),five flavonoids(14-18),one triterpenoid(19),two sterides(20-21),three phthalate esters(22-24)and one alkanol(25)were isolated from the U.rhynchophylloides for the first time.Compounds 1-5 and 8-11 were alkaloids and terpenoids,which were the characteristic components of U.rhynchophylloides,while indole alkaloids have not been isolated.The second chapter introduces the research on the.antihypertensive activity of Uncaria rhynchophylloides How.Including the study of the effects of ET-URH on SHRs blood pressure and myocardial protection,and the screening the inhibitory effects on PE-induced aortal contraction in mice,with some compounds isolated from Uncaria thynchophylloides How.We found Et-URH contains abundant terpenoid and phenolic compounds,which could significantly reduce SHRs blood pressure(P<0.01)and HWI(P<0.05),increase the level of NO(P<0.01)and protect the cardiac tissue of SHRs.The iridoids isolated from U.rhynchophylloides(compound 5)shows a certain inhibitory effect on PE-induced aortal contraction in mice.The third chapter introduces the study of the chemical composition of Bauhinia touranensis.39 compounds,including 3 new compounds and 4 new natural products were isolated and identified from 95%ethanol extracts of the stems and branches of B.touranensis,including one new lactone-dimer(26),one new cyclohexene(33),one new cyanophenol(37),two cyanophenols(38-39),two hydroxyphenylacetates(40-41),six lactones(27-32),three indole alkaloids(34-36),two benzoic acid derivatives(42-43),one chromone(44),eleven flavonoids(45-55),two phenylpropanoids(57-61).All compounds were isolated from this plant for the first time.Compounds 26-41,46,55,61 were isolated from this genus for the first time.The fourth chapter summarized the chemical constituents and pharmacological activities of Bauhinia.So far,225 compounds have been reported from the Bauhinia,mainly were terpenoids and Phenolic compounds,such as flavonoids.Many plants in the genus Bauhinia are also used as traditional Chinese herbs for analgesic,antibacterial,anti-inflammatory,promoting wound healing,immune regulation,antioxidant,anti-tumor,anti-depression and diabetes treatment.
