PART ONEThe isolation,incubation of NSCs from hippocampus of adult SD rats and identification the proliferation and differentiationof NSCsObjective To isolate and incubate neural stem cells from hippocampus of adult SD rats in vitro and to identify its proliferation and differentiation abilities.Methods Neurons in hippocampus of adult SD rats were dissociated mechanically and enzymatically.The suspension culture technique was performed to acquire cell clones.EGF and bFGF were added into the culture in vitro.MTT was adopted for cell growth curve. The expression of nestin ,NSE,GFAP and Brdu(for NSCs,neurons,astrocytes and proliferation of NSCs respectively) were determined by immunofluorescence staining. Results Neural stem cell clones were obtained after primary and passageculture.The proliferation of passage cell clones was increased apparently inthe forth week.The proliferation of the single cell clone was decreased in theeight week.The cell clones from primary ,passage and the single cell culturewere all nestin positive and Brdu positive.The NSE (neuron) and GFAP(astrocytes)were positive after differentiation.
