Haemorrhagic fever with renal syndrome(HFRS) spreads in many countries of Asia, Europe and Africa. The incidence and moutality of the disease is rather high in some areas in China. It is threatening the health and life of the people in our country. Study of HFRS has always been emphasized by scientists of all countries. Owing to emergence and application of monoclonal antibodies (McAbs) against HFRS virus, a great progress has been acheived in the research of HFRS during the past 10 years. At present, many research workers hope that more and better McAbs against HFRS virus have been developed for studies of HFRS in different fields.In this thesis, author established rat hybridoma cell lines secreting McAbs against HFRS virus with rat-rat hybridoma technique and the following several sections are discussed: (1) preparation of anti-HFRS virus rat McAbs; (2) production of anti HFRS V McAbs in vivo or in vitro; (3)purification and application of the rat McAbs; (4)Characterazation and antigenic analysis of the rat McAbs; (5)Measurement of antigenic sites and affinity of the rat McAbs; (6)protection of the rat McAbs for HFRS virus infected mice; (7)development of three kinds of ELISA methods for the detection of HFRS virus specific antibodies in rat sera.I . Preparation of Rat Monoclonal Antibodies against HFRS VirusEleven rat hybridoma cell lines secreting McAbs against HFRS virus were obtained by fusion of rat nonsecreting myeloma cell line(IR983F) with spleen cells from Lou/c-Igk-la rats immunized with HFRS virus strain chen. The growth rate of hybridoma was 37-51%. The positive rate was 39-46%. The McAbs titers in the cell culture supernatant and in the hybridoma ascites detected by a double McAb-ELISA indirect sandwich method were 1×10(?) and 1×10(?)—1×10(?) respectively.
