Objective: To investigate the role of celecoxib in the treatment of autosomal dominant polycystic kidney disease(ADPKD). Methods: In vitro ELISA, Real-time fluorescent quantitative PCRJmmunoihistochemistry and Western blot were used to study cell proliferation,apoptosis and cell cycleinduced by celecoxib.ln vivo examination in serum and urine,pathological evaluation, ELISA.Real-time fluorescent quantitativePCR,RIA,lmmunolhistochemistry and Western blot to study Han:SPRD ratstreated by celecoxib. Results: In vitro PGE2 secretion of cyst lining epithelialcell was reduced by CXB in different concentration and the secretion of EGF and VEGF were reduced by CXB not in a time-dependent manner but in a concentration-dependent manner.lt was demonstrated that the expression level of phospho-MAPK was reduced by CXB. P21CIP/WAF was increased by CXB in a time-dependent manner, and the expression of CyclinA was reduced in the same manner. It can be demonstrated that the cell growth inhibition is probably related to blocking the programmed progression of cell cycle in S stage. CXB can induce apoptosis through activating Caspase3 and decreasing Bcl-2/Bax.ln vivo CXB 3mg/kg and 10mg/kg dosage could decrease...
