Part 1: Establishment of the Acute Rejection and Tolerance Model of Rat Orthotopic LiverTransplantation AIM: To establish an acute rejection and tolerance model of rat orthotopic liver transplantation ( ROLT ) .METHODS: Forty eight ROLT models were established by modified Kamada two-cuff technique, with inbred male DA rats and male LEW rats served as donor and recipient respectively. The recipients were randomly divided into 4 groups: rejection group ( DA→LEW, n=12 ) , tolerance group ( LEW→DA, n=12 ), isogenic group ( DA→DA, n=12 ), and FK506 treatment group [DA→LEW, n=12, lavaged with 0.2 mg/(kg·d) FK506 from 1d to 7d post-transplantation ]. Every 3 recipients from each group sacrificed at 4, 7, 10d post-transplantation, evaluated the changes of serum biochemistry (AST and bilirubin) and the graft histology. The other 6 recipients were left in each group to observe the animal survival time.
