Objective: To investigate the mechanisms of immune escape of hepatocarcinoma cell through Fas/FasL pathway by detecting the expression and the functions of Fas and FasL in human hepatocarcinoma cell line HepG2.2.15; To investigate the reason of low Fas expression in hepatocarcinoma cell by analyzing the distribution of Fas-670 polymorphism; To investigate the inhibit action on T lymphocytes apoptosis induced by hepatocarcinoma cells and the reverse action of Fas and FasL antisense oligodeoxy-nucleotide (ASODN) on the immune escape by transfecting FasL ASODN in hepatocarcinoma cells and transfecting Fas ASODN in Jurkat T lymphocytes. To investigate the effect of cytokine on the apoptosis in hepatocarcinoma cells caused by Interferon- α (IFN- α ) up-regulating Fas expression.Methods: The expression of Fas and FasL of hepatocarcinoma cell line HepG2.2.15 was examined by flow cytometry (FCM). Resistance of hepatocarcinoma cells to Fas-mediated apoptosis was studied using anti-Fas agonistic monoclonal antibody (mAb) CH11.
