Objective: The death rate of tumor patient is high and the life span is short, so the tumor threaten seriously people's health. Along with the development of biological technology, the gene therapy has become the hot spot of present research. To investigate the effect of the c-myc antisense oligodeoxynucleotide on the FHIT gene re-expression gastric cancer cell, and to explore the anti-tumor effection that the oncogene with the anti-oncogene joint action.Methods: The FHIT gene was transfected into human gastric cancer cell line MKN28 by the method of liposome transfection. The FHIT mRNA was detected by RT-PCR and the fhit protein was detected by western-blot. Then, we transfected the c-myc antisense oligodeoxynucleotide into the FHIT re-expression cell transfected with FHIT gene and FHIT gene deficient cell transfected with void vector. The growth inhibition effect was analyzed by MTT while the cell apoptosis was analyzed by AO/EB staining and FCM.Results: After transfected, the FHIT gene and fhit protein were found in MKN28 cell transfected FHIT gene, while in the cell transfected void vector were not. The c-myc antisense oligodeoxynucleotide could inhibit the expression of c-myc protein in MKN28 cell and induce cell apoptosis. The inhibiting growth and inducing apoptosis effect of c-myc antisense oligodeoxynucleotide on FHIT gene re-expression cell was stronger than the effect on FHIT gene deficient cell. Conclusions: The FHIT gene re-expression in the MKN28 cell induces cell apoptosis, and enhances the sensitivity of MKN28 cell to the c-myc antisense oligodeoxynucleotide. The FHIT gene and the c-myc antisense oligodeoxynucleotide joint action plays the formidable anti-tumor effection. These have provided certain experiment foundation for the gene union treatment of tumor.
