Objective:To study the effect of the c-myc antisense oligodeoxynucleotide(asODN) on the proliferation and apoptosis of colon cancer cell SW480 with FHIT gene re-expression.Methods:PRC/CMV vector expressing and empty vector was transfected into human colon cancer SW480 cell by liposome mediation,respectively.Then,c-myc asODN was transfected into SW480 cell. The FHIT and c-myc protein was detected by the method of western blot.The proliferation was assessed by MTT assay.The apoptosis was determined by AO/EB staining and flow cytometry analysis Results:Marked expression of FHIT protein was detected in SW480 cells after transfection with PRC/CMV vector.But it was not observed in SW480 cells after transfection with empty vector.The expression of c-myc protein was significantly inhibited in SW480 cells after transfection with c-myc asODN (p<0.01) .ASODN of c-myc inhited the proliferation and induced apoptosis of SW480 cells.However,the growth-inhibiting and apoptosis-inducing effect were stronger on FHIT+ SW480 cells than FHIT- SW480 cells (p<0.01).Conclusions:The re-expression of tumor suppression gene FHIT and down-regulation of oncogene c-myc haver stronger anti-tumor efficiency.It may be a promising anti-tumor way to interfere symphyseally multiple tumor related gene.
