Polypeptide from Chlamys farreri (PCF, Mr = 879) is a water-soluble polypeptide isolated from Chlamys farreri (a kind of scallop). Studies show that PCF significantly increase the number of immunologic cells exposed to ultraviolet (UV) radiation, the possible role of PCF in the murine thymocytes has not yet been studied. In order to investigate whether PCF has other immunostimulatory effects, we investigated the effects of PCF on thymus in vivo first.The immunosuppressed model was duplicated. The proliferative response of lymphocytes was measured by MTT assay. The percentages of mouse thymocytes subpopulations were analyzed by flow cytometer. We evaluated the effects of PCF against UV radiation by measuring its effects on the murine thymocytes in vitro. The ultraviolet ray irradiation damage models of thymocytes from the Qunming mice was established though orthogonal design .The cells were randomly divided into six groups: control group, ultraviolet ray groups (model, 0.5% PCF, 0.25% PCF, 0.125% PCF, 0.1%VitC). The concentration of GSH-Px, SOD, MDA, T-AOC and the cells'viability were determined by biochemical and MTT methods respectively. The free calcium, mitochondria membrane potential and the rate of apoptosis on thymocytes were tested using flow cytometry (FCM). The expressions of P53, Bax and Bcl-2 proteins were examined by immunocytochemical technica. Also the expression of c-fos mRNA and c-jun mRNA was determined through PT-PCR and the p21, p38 mRNA expressions through the technique of in situ hybridiation. The ultrastructure of the cells was observed through electron microscope. Thymocytes were also pretreated with or without ERKs, JNKs and p38 kinase inhibitors prior to UV irradiation and DNA fragmentation was assessed by agarose gel electrophoresis.
