Pancreatic cancer is highly malignant. Its morbidity is on the increase all over the world. Its prognosis is poor. Nowadays with the development of molecular biology and the finding and cloning of the new oncogene ,anti-oncogene and anti-apoptosis gene related to pancreatic cancer, new targets can be supplied to discuss its pathogenesis, diagnosis and treatment. Of all survivin gene as a member of apoptosis-inhibiting protein gene family, is put more attention to.RNAi is called posttranscriptional gene silencing. As a gene blockage technique, the research of RNAi in gene therapy of tumor have been achieved essential progress recently.Can RNAi play a important role in cure of pancreatic cancer?it is necessary making some based investigates to it.[Objectives] 1st,we will discuss the probability of survivin expression in pancreatic cancer cell strain PANC-1 .2nd,clone the gene from the cell strain PANC-1 and analyze its sequence. 3rd,we will establish the relevant interference vector- shRNA expression vector.4th, observe and detect the inhibition of the survivin expression of the pancreatic cancer cell strain PANC-1 by the interference vector, then choose the high efficient interference vector .5th, detect whether the interference vector can induce apoptosis of the pancreatic cancer cell strain PANC-1.
