AIM: The study of anti-bilharziosis rabbits pulmonary fibrosis of In-vitro cultivated calculus bovis compound preparation through the observation of lung tissue pathological changes of bilharziosis rabbits Praziquantel control group and In-vitro cultivated calculus bovis compound preparation intervention group and expression of FN, LN, c-fos, ERK1,2 in the lung tissue of bilharziosis rabbit Praziquantel control group and In-vitro cultivated calculus bovis compound preparation intervention group to investigate the inhibitory function of In-vitro cultivated calculus bovis compound preparation on the formation of schistosome lung fibrosis and its mechanism, to expand a new metabolic alternative for clinical anti-pulmonary fibrosis .METHODS: Observe the morphological changes of lung tissue in 20 cases of bilharziosis rabbits Praziquantel control groups and 20 cases of In-vitro cultivated calculus bovis compound preparation interventional groups by light microscope and electron microscope; detect the expression level of FN, LN , c-fos in the lung tissue of 20 cases of bilharziosis rabbits Praziquantel control groups and 20 cases of In-vitro cultivated calculus bovis compound preparation interventional groups by immunohistochemical method; detect the expression of ERK1/2 in the lung tissue of 20 cases of bilharziosis rabbits Praziquantel control groups and 20 cases of In-vitro cultivated calculus bovis compound preparation interventional group by Western Blot tech.RESULTS: more alveolus exudate in the lung tissue of bilharziosis rabbits Praziquantel control group under light microscope, the alveolar space is fairly dry in the lung tissue of In-vitro cultivated calculus bovis compound preparation interventional group; more alveolus exudate in the lung tissue of bilharziosis rabbits Praziquantel control group under electron microscope, can be seen more macrophage, alveolus angiotelectasis, the structure of capillary three-tier barrier is veil, the alveolus interspace fiber is little increased in parts of the subregion of the lung tissue of In-vitro cultivated calculus bovis compound preparation interventional group, the structure of I type epithelium, basal lamina, endodermis is fundamentally integrity, there is no manifest exudation from the alveolar space, the vessels have no congestion; the expression of FN, LN, c-fos is positve or strong positive in the lung tissue of the bilharziosis rabbit Praziquantel control group the expression of FN, LN, c-fos is negative or weak positive in the lung tissue of In-vitro cultivated calculus bovis compound preparation interventional group ; the expressional intensity of ERK1/2 in the lung tissue of the control group is manifestly higher than in the lung tissue of In-vitro cultivated calculus bovis compound preparation interventional group (P < 0.01CONCLUSION: In-vitro cultivated calculus bovis compound preparation can effectively prevent biharzia worm lung fibrosis, cut down the content of the extracellular matrix; the inhibitory function of In-vitro cultivated calculus bovis compound preparation is possibly through improving lung microcirculation. Section I The Effects of In-vitro Cultiated Calculus BovisCompound Preparation(ICCBco) On Pathologic Morphology In Experimental Pulmonary Fibrosis In Rabbits With Schistosomia JaponicumObjective To investigate the effect of in-virto cultivated calculus boviscompound preparation(ICCBco) on pathologic morphology of pulmonary tissue under the process of pulmonary fibrsis induced by schistosomia japonicum in rabbits. Methods Observe the morpholobical changes of lung tissue in 20 casesof bilharziosis rabbits Praziquantel control groups and 20 cases of In-vitro cultivated calculus bovis compound preparation interventional groups by light microscope and electron microscope. Results more alveolus exudate in the lung tissue of bilharziosis rabbitsPraziquantel control group under light microscope, the alveolar space is fairly dry in the lung tissue of In-vitro cultivated calculus bovis compound preparation interventional group; more alveolus exudate in the lung tissue of bilharziosis rabbits Praziquantel control group under electron microscope, can be seen more macrophage, alveolus angiotelectasis, the structure of capillary three-tier barrier is veil, the alveolus interspace fiber is little increased in parts of the subregion of the lung tissue of In-vitro cultivated calculus bovis compound preparation interventional group, the structure of I type epithelium, basal lamina, endodermis is fundamentally integrity, there is no manifest exudation from the alveolar space, the vessels have no congestion. Conclusions ICCBco can exert significant effect on improvingregeneration and damage of pulmonary cytes, M eanwhile effectively protect liver from deposition of collagenous fiber and has inhibitory effects on pulmonary fibrosis. Section II The Effects of In-vitro Cultiated Calculus Bovis Compound Preparation(ICCBco) On expression of extracellular matrix and collagen in experimental pulmonary fibrosis in rabbits with schistosomia japonicumObjective To investigate the effect of in-vitro cultivated calculus bovis compound preparation(ICCBco) on expression of fibronectin and laminin and collagen in experimental pulmonary fibrosis in rabbits with schistosomia japonicum. Methods Detect the expression level of FN and LN in the lung tissue of 20cases of bilharziosis rabbits Praziquantel control groups and 20 cases of In-vitro cultivated calculus bovis compound preparation interventional groups by immunohistochemical method. Observe the collagen changes of lung tissue in 20 cases of bilharziosis rabbits Praziquantel control groups and 20 cases of In-vitro cultivated calculus bovis compound preparation interventional groups by Mallory technology. Results The expression of FN and LN is positve or strong positive in thelung tissue of the bilharziosis rabbit Praziquantel control group the expression of FN and LN is negative or weak positive in the lung tissue of In-vitro cultivated calculus bovis compound preparation interventional group. more collagen in the lung tissue of bilharziosis rabbits Praziquantel control group under light microscope, less collagen in the lung tissue of In-vitro cultivated calculus bovis compound preparation interventional group; Conclusions The result indicated that the ICCBco might effectively protect pulmonary in rabbit with schistosomia japonicum by way of inhibiting expression of extracellular matrix. Section III The Effects of In-vitro Cultiated Calculus Bovis Compound Preparation(ICCBco)On expression of ERK1/2 /c-fos in experimental pulmonary fibrosis in rabbits with schistosomia japonicumObjective To investigate the effect of in-vitro cultivated calculus bovis compound preparation(ICCBco) on expression of ERK1/2 /c-fos in experimental pulmonary fibrosis in rabbits with schistosomia japonicum. Methods Detect the expression level of c-fos in the lung tissue of 20 cases of bilharziosis rabbits Praziquantel control groups and 20 cases of In-vitro cultivated calculus bovis compound preparation interventional groups by immunohistochemical method. Detect the expression level of ERK1/2 in the lung tissue of 20 cases of bilharziosis rabbits Praziquantel control groups and 20 cases of In-vitro cultivated calculus bovis compound preparation interventional groups by western blot method. Results The expression of ERK1/2 and c-fos is positive or strong positive in the lung tissue of the bilharziosis rabbit Praziquantel control group the expression of ERK1/2 and c-fos is negative or weak positive in the lung tissue of In-vitro cultivated calculus bovis compound preparation interventional group. Conclusions The result indicated that the ICCBco might effectively protect pulmonary in rabbit with schistosomia japonicum by way of inhibiting expression of ERK1/2/c-fos.
