The Effect On The Activity Of Ion Channels Of S₁₈₀ Tumor Mice Erythrocyte Membrane Of Ffructus Schisandrae Chinensis Polysaccharides

Objectives:The article was begin from the abstraction decontamination and identification Of Fructus Schisandrae Chinensis polysaccharides,Inhibition tumor experiment of Fructus Schisandrae Chinensis polysaccharides were done ,Based on this study , Study on the activity of the ion channels of the erythrocyte membrane S₁₈₀ mice to discuss antitumor mexhanisms in terms of the erythrocyte membrane fuction ,and provid redical foundation for the clinic application.Methods: the abstraction Fructus Schisandrae Chinensis polysaccharides using ethyl alcohol; Examined the content of Fructus Schisandrae Chinensis polysaccharides using phenol-sulfuric acid ;Analysis and indentition of Fructus Schisandrae Chinensis polysaccharides were done using HPCE.Spectrophotometer method were used to measure the activities of Na⁺- K⁺- ATPase ,Ca²⁺-Mg²⁺-ATPase and Band3 protein,to measure the sialic acid content, the degree of closure of erythrocytes membrane and the fluidity of the cell membrane;Examined the expression of Band3 protein using SDS-PAGE and Weston blot;Examined the change of [Ca²⁺] using Laser scanning confocal microscopy ;Examined the natural ability of adhering turner cells of erythrocytes by rosette formation test.Results and conclusions:(1) Fructus Schisandrae Chinensis polysaccharides can enhance the activities of Na⁺,K⁺-ATPase, Ca²⁺,Mg²⁺-ATPase and Band3 protein on erythrocytes of the S₁₈₀ tumor mice ( P < 0. 01) ;(2) Fructus Schisandrae Chinensis polysaccha- rides can increase the fluidity of the erythrocyte membrane of the S₁₈₀ tumor mice,the sialic acid content on the erythrocyte membrane and the degree of closure of tumor-bearing mice (P<0. 01) , decrease the [Ca²⁺] in tumor-bearing mice (P <0. 05) ;(3) Fructus Schisandrae Chinensis poly- saccharides can increase the natural ability of adhering the tumor cells by erythrocytes (P<0. 01) ,the immunization ressette ratio is 24.17 percent;(4)the content of Fructus Schisandrae Chinensis polysaccharides is 85.56 percent,it has Rha,Gal,Glu,Ara and two unknown monosaccharides. (5)The inhibit-tumor ratio of Fructus Schisandrae Chinensis polysaccharides is 45.4 percent.For the above mentioned , Fructus Schisandrae Chinensis polysaccharides can increase the activities of ion channels, found the interpromomtion relationship among the ion channels adjusting and controling the ion balance, immprove the structure of RBC membrane and enhance the function of immunity of erythrocytes ,to anti-tumor.
