Objective:To explore the effect and mechanism of VEGF and Ang-1/Ang-2 on differentiation and maturation of choroidal neovascluarization.Methods:RUVECs were cultured in vitro scratch and MTT assay were applied to detect the effects, including the ability of migration and proliferation, as well as the changes of microcellular structure of VEGF,Ang-1/Ang-2 group. Mice of CNV model were received intravitreous injection with VEGF or Ang-1, the FFA images and pathological section were analyzed to detect the effects of factor Ang-1 or VEGF.Results:In scratch and MTT assay, VEGF was found no significant improvement ability to the migration and proliferation of RUVEC. So to Ang-1 and Ang-2. Combination of VEGF+Ang-1 or VEGF+Ang-2 could significantly improvement the migration and proliferation of RUVEC. In CNV model mice Ang-1 group was found less later period leakage than the control group, while in VEGF group is more than the control group. The scar was found in pathological tissue section of Ang-1 group, and granulation tissue was found in VEGF and control group.Conclusions:Single factor of VEGF or Ang-1/Ang-2 cant effect the migration and proliferation of RUVEC. While combination of VEGF+ Ang-1 or VEGF+ Ang-2 were proved effective. Ang-1 can suppress the formation and maturation of CNV and improve the scar formation.
