Objective:To explore the effects of VEGF,Ang-1/Ang-2 and TCM "Liangxuehuayu" on maturation of choroidal neovascluarization in vitro and in vivoMethods:RUVEC were cultured in vitro. MTT assay was applied to observe the proliferation of RUVEC affected by VEGF,Ang-1/Ang-2. The surface and internal structure of RUVEC were observed by transmission electron microscopy and scanning electron microscopy. In vivo test, CNV rat model were created to compare the different effect of intravitreous injection with Ang-1 and stomach medicine with TCM.Results:In MTT assay, VEGF can improve the proliferation of RUVEC significantly in some concentration scope. Combination of VEGF and Ang-1 can significantly improve the proliferation of RUVEC. The cell junctions were much more in VEGF/Ang-1 group than single factor of VEGF. The cell functions were in vigorous activities by the effect of VEGF and Ang-1. In vivo test,the intravitreous injection with Ang-1 has similar effect on reducing vascular leakage with stomach medicine with TCM.Conclusions:Combination of VEGF and Ang-1 can induce maturation of choroidal neovascluarization and provide new theoretical method and experimental evidence for the therapy of choroidal neovascluarization. TCM"Liangxuehuayu"has good inhibition effect on CNV animal model.
