Objective:1. To Discusses the rationality and feasibility of using the theory and means of epigenetics to TCM research.2. To investigate microcosmic essentials of kidney deficiency with blood stasis syndrome at the gene level in diabetes.Method:1. Compare the concept and content of epigenetics with TCM genetics to get syndrome study need to introduce the theory and means of epigenetics.2. Clinical investigations with scaling tables on four diagnosis, kidney-deficiency, and blood stasis.3. mRNA and miRNNA microarray tests to screen out potential differentially expressed genes for type 2 diabetes with kidney-deficiency and blood stasis syndrome.Results:1. With applying the concepts and methods of epigenetics to the field of Chinese medicine research, it will provide new ideas, new methods and new technology in many aspects such as viscera state heredity, endowment, combination of disease and syndrome, and lay the foundation for prevention and treatment of disease.2. A pair of diabetes twins and their families were systematically investigated during the last consecutive two year periods to suggest kidney-deficiency and blood stasis syndrome hasing a hereditary basis.3. Some metabolism, transcriptional regulation and immun genes were screened out which may play important roles in type 2 diabetes with kidney deficiency and blood stasis syndrome.4. Several candidate differentially expressed miRNA were screened out and verified by differentially expressed mRNA.Conclusion:Chinese medicine is elaborated from a macro up the body's physiological and pathological processes, TCM syndrome, efficacy, genetic medicine, and many other problems are difficult to be explained by classical simple single-gene mutations, which need to introduce the theory and means of epigenetics research.Through investigate of a pair of diabetes twins and their families, suggest diabetes is connected with kidney deficiency and blood stasis syndrome. Use of mRNA and miRNNA microarray, some metabolism, transcriptional regulation and immun genes were screened out which may play important roles in type 2 diabetes with kidney deficiency and blood stasis syndrome, and several candidate differentially expressed miRNA were screened out and miR-32 were verified may play important roles in type 2 diabetes.
