Objectives To investigate expression of TIP30 and OPN,as well as relationship between expression and prognosis in the largngeal squamous cell carcinoma (LSCC) tissues, and to explore the biological effect and possible mechanism of inhibiting tumor metastasis by TIP30 on laryngeal cancer. Methods SP immunohistochemical method was used to study the TIP30 and OPN expression on sections of surgical operated specimens of 105 laryngeal squamous cell carcinomas and adjacent tissues. Clinicopathological features were anlysized. Interferenced or introduced TIP30 into Hep-2 cells by Lentivirus vector, then OPN, VEGF and MMP-9 expressions were detected. TIP30 was detected the inhibition of proliferation, migration and invasiveness of Hep-2 cells in vitro. Construction laryngeal tumor model and observe the tumorigenesis and morphological changes. The different expressions of OPN, VEGF and MMP9 were observed by western blotting assay. The microvessel density was examined by Immunohistochemical method. Results There is an inverse correlation in protein levels of TIP30 and OPN from analysis of LSCC tissues. The TIP30 expression levels were significantly correlated with tumor stage, grade and the presence of lymph node, not correlated with age, gender and tumor primary site. LSCC patients with lower expression level of TIP30 have a higher recurrence rate and poorer prognosis (P=0.010, P=0.03, respectively). Introduction of TIP30 in Hep-2 cells significantly reduced cell growth, migration, invasion and metastasis, as well as decreasing expressions of VEGF and MMP-9 by OPN. TIP30 played a role in inhibiton of tumor growth and metastasis by downgulation of OPN to decrease VEGF and MMP-9 expressions. Conclusions Down-regulation of TIP30 expression is correlated with the poor prognosis in patients with LSCC. The expression of TIP30 is inverse correlation with that of OPN. TIP30 maybe participated in the development and metastasis of LSCC. TIP30 reduces MMP9 and VEGF expressions by OPN signal pathway and inversely regulates cell survival and invasion in vivo or vitro, hence it could serve as an important tumor metastasis suppressor gene, which is probably expected to provide a theoretical basis for treatment in largngeal carcinoma.
