Objective To detect the expression of osteopontin (OPN) and urokinase-type plasminogen activator(uPA) in squamous cell carcinoma tissue and adjacent normal tissue of human larynx.In this study, the contrast analysis from the different clinic features of the tumors was carried out, and the significance of OPN and uPA in histogenesis and metastasis of laryngeal carcinoma was investigated.The objective was to supply the correlated theory to judging the biological behaviors and prognosis of the tumors.Methods 64 specimens of laryngeal squamous cell carcinoma and 18 specimens of adjacent normal tissue were selectd in the experiment.The expression of OPN and uPA in laryngeal carcinoma tissue and adjacent normal tissue was detected using immunohistochemistry method. To count the numbers of OPN and uPA,which were positive expression. The statistical analysis was used to reveal the difference of OPN and uPA respectively from the different aspects of tumor. The correlation between OPN and uPA in laryngeal squamous cell carcinoma was analyzed by Spearman correlation analysis.Results In 64 specimens of laryngeal carcinoma tissue the positive expression rates of OPN and uPA were 62.5%(40/64),65.6% (42/64) respectively ,which were higher obviously than in adjacent normal tissue(P<0.05).In stage III to stage IV laryngeal carcinoma tissue, the positive expression of OPN and uPA were 73.8% and 76.2% respectively,which were higher obviously than in stage I to stage II (P<0.05).Furthermore, the expression rates of OPN and uPA in speciemns which had metastasized the lymph nodes were higher than the ones which had not metastasized the lymph nodes(P<0.05).The expression of OPN and uPA in carcinoma tissue was related to clinical pathological stage and lymphatic metastasis(P <0.05), but not related to age, sex or differentiation degree (P>0.05). The expression of OPN was positively correlated with that of uPA in laryngeal carcinoma tissue (r =0.6625, P<0. 05).Conclusion In laryngeal squamous cell carcinoma,The expression of OPN and uPA was related to clinical pathological stage and lymphatic metastasis,and they had synergism.The expression of OPN and uPA could reflect the biological characteristics of laryngeal carcinoma and be used as important moleculer marker in prognosis of laryngeal carcinoma.
