Similar to other malignant tumors, the undesirable prognosis in the patients with laryngeal squamous cell carcinomas ( LSCC) has been attributed to cervical lymph node metastasis. In LSCC, lymph node metastasis the most important event in the failure of the treatment of LSCC. Therefore, it is a major goal to identify the molecular mechanisms of invasion and metastasis of the LSCC.Metastasis is a complex series of events that involves several gene products, the MTA1 gene is a recently identified novel candidate metastasis - associated gene. Previous study showed the mRNA expression levels of MTA1 correlated with metastatic potential in several human neoplasms.Gene expression of nm23 has been investigated in number of tumors, including breast cancer and others. Some people reported that nm23H1 down - regulation was associated with metastasis in some tumors. But others reported the contrary result. Thus the significance of nm23H1 in the metastatic cascade remains unknown.We also calculated the MTA1/nm23H1 ratio of the OD in each case to value the relation between the ratio and the metastasis.We applied RT - PCR technique to detect the MTA1 And nm23H1 expression at molecule level. RT - PCR technique is sensi-live and specific in analysis and estimation of tumor micrometastasis by detecting the molecules interrelated with tumors.Experimental MaterialsThe 48 sample of laryngeal carcinoma and 16 samples of normal tissue were acquired from the patients hospitalizing and underwent operation in the ENT department of the first clinical Medical University between September, 2001 to February , 2002 . Of them , there were 16 supraglotti laryngeal carcinomas patients with cervical lymph node metastasis (the first group) , 16 supraglotti laryngeal carcinomas patients with cervical lymph node - negative metastasis ( the second group) , 16 glotti carcinoma with cervical lymph node - negative metastasis (the third group) and 16normal tissue (the fourth group). All of the patients had not received either chemotherapy or radiotherapy before surgery. The pathological results were all squamous cell carcinoma. The samples were resected and snap ?frozen in liquid nitrogen , immediately after surgical resection . Specimens were stored at -Experimental MethodsWe applied RT - PCR technique to detect the MTA1 and nm23H1 expression at level of molecule. The relative amounts of MTA1 and nm23H1 expression were normalized to B - actin expression , and the MTA1 and nm23H1 expression in the laryngeal carcinomas with cervical lymph node metastasis was compared with that in the corresponding laryngeal carcinomas with cervical lymph node - nega-live metastasis and normal mucosa. We used the SPSS 10.0 statistical software system ( SPSS Institute, Inc) to do the calculations . P < 0. 05 were considered to be statistically significant .ResultThe expression of MTAlmRNA by RT - PCR in the first group, 16 specimens was positive with 100% positive rate, in the other groups, none was positive with 0% positive rate.The result of RT -PCR for nm23Hl showed that the relative ab-sorbance was a mean value of 1. 633 ( standard deviation, 0. 241) in the cases of the first group, a mean value of 1. 502 ( standard deviation , 0.298 ) in the cases of the second group, a mean value of 1.476 (standard deviation, 0. 339) in the cases of the third group and a mean value of 1.997 ( standard deviation, 0. 618) in the fourth group. The difference between the first and the second group, the first and third group were not statistically significant ( P > 0. 05 ) . But the difference between the first and the fourth group were statistically significant ( P <0.05 ) . The relative absorbance of the nm23Hl in the 32 laryngeal carcinoma of the cervical lymph node - negative metastasis (the fifth group) was a mean value of 1.489(standard deviation, 0. 296 ).
