objective:To observe the effect of XiongZhen Cream in rat cutaneous incised wound healing and investigate the mechanism of accelerating the rat back wound healing.Methods:After setting up the model, the rats were randomly assigned to 4 groups: model group, Beijifu group (western medicine control group), Xiongzhen Cream group, Beijifu + Xiongzhen Cream group. The medicine were attach to the skin twice a day. After setting up models for 3,7 and 14 days, the rats were killed to perform immunohistochemistry study. S-P method of immunohistochemistry was used to detect CD34, number of fibroblasts and CD68 macrophages of the local granulation tissue. Double sandwich ELISA was used to detect the level of VEGF and CRP in the plasma. We used image analysis equipment and computer software to analyze the image of each group. The data were expressed as Mean±SD and was analyzed by SPSS 18.0.Results: The Xiongzhen cream significantly improved the weight of the rats after setting up model, and it was significantly better than rats in model group (P<0.01). The area intensity, number intensity and perimeter intensity of granulation tissue were significantly better in Xiongzhen cream group than model group in all three time points(P< 0.01). The improvement of number of fibroblasts was significantly better in Xiongzhen cream group than model group in all three time points. Xiongzhen cream significantly lowered the level of VEGF in rats'plasma, and it was significantly better than model group (P< 0.01). Xiongzhen cream significantly lowered the level of CRP and TNF-a in rats' plasma (P<0.01 or P< 0.05). Xiongzhen cream significantly improved the level of macrophages in granulation tissue in 7th day, and it was significantly better than model group(P< 0. 01).Conelusion:The Xiongzhen cream accelerate the healing of skin trauma of the rats and improved the quality of healing. This might be related to improvement of microvessels and number of fibroblasts of granulation tissue and improvement of local immune, expression, reduce of inflammation in the granulation tissue.
