Sinisan comes from Treatise on Febrile and Miscellaneous Diseases writed by Zhongjing Zhang in Han Dynasty. It is made up of radix bupleuri, Chinese herbaceous peony, citrus aurantium and Radix liquirtiae. It treats diseases with such syndromes caused by impediment in the functional activities of Qi .It is representative recipe to regulate the functional activities of Qi through development of later medical specialist. It apply to such syndromes caused by impediment in the functional activities of Qi as diseases caused by external factors or internal injury. We have made good therapeutic results by using variant sinisan to treat ulcerative colitis (UC) of liver-depressed rats. The main function of variant sinisan is to regulate the functional activities of Qi with clearing heat, clearing damp and invigorating the circulation of blood. To approach functional mechanism of variant sinisan to treat UC of liver-depressed rats, we have made the following experimental study.Objective: To approach functional mechanism of variant sinisan to treat UC of liver- depressed rats model by observing effect of variant sinisan to treat it and influence of Oxygen Free Radicals(OFR) damage and inflammatory cytokines.Methods: The UC of liver-depressed rats model was induced by TNBS and ties, and divided 6 groups: normal group; model group; SASP treatment group;3 variant sinisan treatment group included large dose group, medium and little dose group. The effect of variant sinisan to treat UC of liver-depressed rats is observed from integrity, tissue, cell and molecule level. The following is the main content: the effect of variant sinisan to treat UC of liver-depressed rats; the effect of variant sinisan to OFR damage of UC of liver-depressed rats; The effect of variant sinisan to inflammatory cytokines content and mRNA of UC of liver-depressed rats.Results: observation in optical microscope: there was many colitis part which show acute and chronic characteristic in colon in model group, variant sinisan group similar to normal group. MPO content have a distinct increase(P<0.01) in model group than in normal group. Compared to model group, MPO have a distinct decrease(P<0.01) in 3 variant sinisan group. MDA and Interleukin-1β and Interleukin-6 content with a distinct decrease, SOD content with a distinct increase in variant sinisan group are significant (P<0.01)compared to model group. In 3 variant sinisan group, the therapeutic results of medium dose group is better than the other two and is no significant(P>0.05) compare to normal group. The therapeutic results of variant sinisan group of large and medium dose better than that of SASP treatment group(P<0.05).Compared the therapeutic results of variant sinisan group of little dose with that of SASP treatment group ,there is no significant(P>0.05).So is gene expression of Interleukin-1β.Conclusions: Variant sinisan has good therapeutic results to UC of liver-depressed rats. The therapeutic results of medium dose group is best in all groups, which conforms to the TCM theory that Chinese medicine dose should be a small amount in treating disease with relievingthe depressed liver and regulating the functional activities of Qi. The functional mechanism of variant sinisan may be connected with inhabiting the increased inflammatory cytokines such as Interleukin-1β and interleukin-6 and the expression of Interleukin-1βmRNA, rectifying abnormal immune function and lower reaction of immune cell to inflammation, which benefit to dispel inflammation and repair tissue. At the same time, the experiment provides foundation for treating UC of liver depression in clinic.
