| Objective: Probing into the differential diagnosis and treatment of YIN huang jaundice.To study the pathogenesis of YIN huang jaundice, and reveal the mechanisms of warming YANG and promoting blood flow and eliminating jaundice Fang(WYAPBFAEJF) for YIN huang jaundice.Methods: 1.The theories research adopts method towards bibliographic retrieval. 2. The experiment study:Wistar rats were randomly divided into 6 groups: the normal control group , YIN huang jaundice model group ,YANG huang jaundice model group, WYAPBFAEJF high-dosage treatment group, WYAPBFAEJF low- dosage treatment group,control treatment group. 3. The changes of liver function and hepatic pathology and ultrastructure were observed respectively in each group. 4. The RIA assay was used to detect Cortisol(Cor)>, ACTFL TXB2 and 6-Keto-PGF, a in plasma of rats . 5. The TUNEL assay and the immunohistochemistry assay were used to detect the apoptosis of hepatocytes and the expression of Bcl-2,Bax in hepatocytes respectively. 6. ELISA assay and in site hybridization technique were used to detect the expression of IL-4 and IFN- Y and PKC a mRNA respectively in spleen cells of rats. 7.To prepare the WYAPBFAEJF drugs'serum ,it effected Jurkat cells ,then the ELISA assay was used to detected the secretion of IL-2 and IFN - Y in Jurkat cells,and take the Flow Cytometry to detect the IL-2R expression on Jurkat cell surface .Results: l.The YIN huang jaundice was put down in writing in General Treatise on the Causes and Symptoms of Diseases by Chao Yuanfang in the Sui Dynasty. The differential diagnosis and treatment system of YIN huang jaundice was put forward by Han Zhihe in the Song Dynasty. The insufficience of the spleen and cold-damp and blood stasis was the fundmendtal pathogenesis of YIN huang jaundice. 2. Model rats had obvious anomaly in function and histology and ultrastructure in liver. WYAPBFAEJF could improve the impairment level of liver .3. The level of TXB2 in plasma increased significantly ,the level of 6-Keto-PGF| a decreased; The content of Cor and ACTH in YIN huang jaundice rats was showing a tendency to increase,but there were significance on the statistics. WYAPBFAEJF could reduce the level of TXB2 and the odds of 6-Keto-PGFi a . 4. The rate of hepatocyte apoptosis was increased significantly, the expression of bcl-2 in hepatocyte decreased greatly,the expression of Bax in hepatocyte increased greatly in YIN huang jaundice model rats. WYAPBFAEJF could increase the expression of Bcl-2 in hepatocyte,decrease the expression of Bax in hepatocyte. 5. Model rats have obvious anomaly in specific value of IFN-Y /IL-4,the expression of PKC a gene in spleen cells decreased greatly.But WYAPBFAEJF could increase the level of IFN- Y and the expression of PKC a gene in spleen cells. 6. WYAPBFAEJF serum could obviously add the secretion of IL-2 and IFN- Y in Jurkat cells, but the secretion of IL-2 could add as theconcentration drugs'serum increase, the secretion of IFN- Y could decreased as the concentration drugs'serum increase,and the expression of IL-2R on Jurkat cell surface could decreased as the concentration drugs'serum increase.Conclusion: 1.The study has suggested that the insufficience of the spleen and cold-damp and blood stasis is the fundmental pathogenesis of YIN huang jaundice. 2.By the means of the experimental study,we detected that the YIN jaundice rats have obvious anomaly in function and histology and ultrastructure in Hver.S.The results suggest that the great amount rate of hepatocyte apoptosis may be the pathogenesis of YIN huang jaundice.4.The HPA function have not chance significantly ,the blood stasis is one of it's essential pathogenesis.5.The expression of Thj/Tha cytokines of spleen cells show disorder ,it related to the expression of PKC a mRNA.6. The WYPBFEJF can amend the changes of hepatic pathology and ultrastructure in YIN huang jaundice rats, we detect that it have truck with preventing hepatocyte apoptosis perhaps by adjusting the expression...
