Objective Probing into the Traditional Chinese Medical Pathologensis of benign prostatic hyperplasia .To study the relationship of cell signal factors and benign prostatic hyperplasia, clarify the mechanism of Xian Jia prescription restraining benign prostatic hyperplasia.Methods (1)S.D male rats were divided into 6 groups :normal group, model group, Long Bi Shu group, Xian Jia prescription low dose group, middle dose group and high dose group.Rats were castrated , then subcutaneous injected testosterone propionate for 30 days.(2)Observe the changes of prostate weight , prostate exponent, histology under light microscope of each group.(3)Take the radioimmunochemistry to measure the thromboxane B2,6-Keto-prostaglandin F1a content of prostate constitution.(4)Using the radioimmunochemistry to measure LPO, SOD content of prostate constitution. (5)Use the method of immunohistochemistry to detect the expression of FAS, Proliferating cell nuclear antigen (PCNA) in each prostate constitution.(6)Take the radioimmunochemistry to measure the IGF-1, IGFBP-3 content of prostate constitution.Results (1)The protate weight, epithelia cell height increased significantly ( P<0.01 ), Xian Jia prescription can restaint the changes. (2)In histology the model rats have obvious anomaly, Xian Jia prescription can relieve the anomaly.(3)The rate of TXB2/6-Keto-PGF1a in model rats increased greatly ( P<0.01 ), Xian Jia prescription can low the level . (4)The rate of LPO/SOD in model prostate cells increased significantly, Xian Jia prescription can adjust LPO/SOD.(5)The expression of PCNA in model prostate cells increased greatly , but the FAS decreased significantly, Xian Jia prescription can low the PCNA expression and increase the FAS expression.(6)The IGF-1 , IGFBP-3 content of prostate constitution in model rats increased significantly ( P<0.01 ), Xian Jia prescription can low them.Conclusion The study has suggested that the "deficiency of Kidney and excess of liver" is the fundmental pathogenesis of benign prostatic hyperplasia.By the experimental study , we detected the TXB2/6-Keto-PGF1a, LPO/SOD rate and IGF-1, IGFBP-3 content of prostate constitution in model rats increased, the expression FAS reduced and PCNA increased in model group.The results suggest these result in benign prostatic hyperplasia. Xian Jia prescription can significantly control weight ofrats' prostate , we presume Xian Jia prescription can regulate the cell factors , induce the cell apoptosis of benign prostatic hyperplasia.So, benign prostatic hyperplasia was restrained.
