Ischemia cerebral vascular disease is a serious disease which still threatens human health and life quality. Now it is generally agreed that apoptosis is the chief death pattern of neurons in penumbra after ischemia and reperfusion caused injury in the brain. To contain apoptosis can reduce the injury of nerve cells in edema band. The Chinese medicinal treatment is predominant in this stage. In this thesis, the protective effects on the brain and the mechanism of fufangbaihuqianhu(FFBHQH) were studied through the experiments on animals and cell culture experiments guided by Chinese medicinal theory and based on the neuron apoptosis technique. l.The protection of FFBHQH to MCAO/R experimental rats The 4-0 type surgical nylon suture of Approximately 20 mm (with its tip rounded by heating near a flame) was inserted from the left external carotid artery and put into the internal carotid artery until it blocked the start-point of the MCA for 120 min. Male Wistar rats, weighing 240-300g, were divided into the five groups : normal, control, FFBHQH (high, middle, low dosage). FFBHQH were administrated by p.o, and all of the experimental indicators were observed and recorded on the 1st 2nd, 3rd day.1.1 The effect of FFBHQH on the formation of brain infarction and nerves function of nerves when transient focal cerebral ischemia/ reperfusion happened72h after MCAO/R surgery, brain tissues were cut into thick slices and stained with TTC, the datas were analysed by computer-assisting image analysis system. Results showed that brain infarction in the groups FFBHQH decreased (P<0.01), especially in the high-dosage group FFBHQH. The symptoms of the injured nerves function in the three dosage groups of FFBHQH have improved significantly (P<0.05 or P<0.01).1.2 The effect of FFBHQH on the morphological changes of brain tissues of MACO/R experimental ratsThe brain tissue slices were stained with HE. The control group it was found that the neuronal morphologic changes included numerous necrosis of nerve cells and infiltration of inflammatory cells in the infarction area and Neuron cells have lost partly, the remained cells swollen, the acidophilia change of cytoplasm, and there was glial cell reaction in hippocampal CA1 area. These changes in the three dosage groups FFBHQH were smaller than those in control group on the 3rd day. These results indicated that FFBHQH could improve the absorption of infarction and relieve the cerebral infarction.2.The effect of FFBHQH on nerve cell apoptosis of MCAO/R experimental ratsThe adjacent sections were stained with the terminal deoxynucleotidyltransferasemediated dUTP nick end-labeling (TUNEL) method. The apoptosis rates ware measured with microscopic analysis (400 field) by counting the positive cells in 1000 cells. Results showed that the positive TUNEL stained cells increased notably in the control group. Apoptosis cells could be found in from the center of infarction to the cerebral edema zone. The average apoptosis ratios were 115.6 8.31, on the 3rd day, while those in groups FFBHQH were 99.2% , 65.2%, 63.2(P<0.01, P<0.05), which suggested that FFBHQH could restrain nerve cells apoptosis.3. The effect of FFBHQH on the apoptosis correlated gene Bcl- 2 , Bax, Caspase - 3 protein expression with nerve cell of MCAO/R experimental rats.Immunohistochemistric technology was used to examine gene Bcl-2, Bax, Caspase-3 protein expression in the brain tissue slices. Compared with the normal group, the cells with Bcl-2 positively in controlgroup increased significantly on the 3rd day(P<0.05). Compared with the model group, The positive expression of Bcl-2 increased in FFBHQH group on the 3rd day (P<0. 05 or P<0. 01). Compared with the normal group, the cells with Bax , Caspase-3 stained positively in the control group increased significantly on the 3rd day( P<0. 01), Compared with group model, it increased in the FFBHQH groups on the 3rd day (P<0.01 or P<0.05).It was indicated that the mechanism of restraining apoptosis was related with the adjusti...
