Purpose:By studying the expression of PPARγ2 mRNA and protein in bone, kidney and hypothalamus of osteoporosis rats caused by ovarian remove,to analyze and evaluate the relation of the pathomechanism and the signal transduction of PPARγ2,to explore the Chinese herbs with marrow-supplying,bone-increasing and kidney-tonifying(in this experiment refer to kidney-tonifying Chinese herbs) combat osteoporosis,one of the mechanisms may be the regulation of signal transduction PPARγ2.Materials and Method:Experimental rats were made by cutting ovarian,then rats were divided randomly into eight groups:normal group,sham-operated group,model control group,kidney-tonifying Chinese herbs low dose group, kidney-tonifying Chinese herbs high dose group,tonifying spleen Chinese herbs the control group(Tonifying the Middle and Replenisling Qi Decoction),Gaitianli the calcium tablet control group(oyster calcium),Gushukang particle control group.Fed the rats one week later.12 weeks later,the expression of PPAγ2 mRNA and protein in bone,kidney and hypothalamus were,measured with RT-PCR and Western Blotting.Results:1.The results of RT-PCR:the expression of PPARγ2 mRNA of bone,kidney and hypothalamus existed in normal group.When compared with normal group,the level of the expression of bone,kidney and hypothalamus of PPARγ2 mRNA of sham-operated group has no markedly discrepancy(P>0.05),the level of the expression of bone and kidney of model control group was obviously increased(P<0.01),the expression of hypothalamus was obviously decreased (P<0.01).12 weeks later,the expression of PPARγ2 mRNA in bone and kidney of. every treating group were obviously decreased(P<0.01),when compared with model control group,the expression of hypothalamus was obviously increased(P<0.01). Kidney-tonifying Chinese herbs high dose group was better than the others(P<0.01).2.The results of Western Blotting:the expression of PPARγ2 protein of bone, kidney and hypothalamus existed in normal group.When compared with normal group,the level of the expression of bone,kidney and hypothalamus of PPARγ2 protein of sham-operated group has no markedly discrepancy(P>0.05),the level of the expression of bone and kidney of model control group was obviously increased(P<0.01),the expression of hypothalamus was obviously decreased (P<0.01).12 weeks later,the expression of PPARγ2 protein in bone and kidney of every treating group were obviously decreased(P<0.01),when compared with model control group,the expression of hypothalamus was obviously increased (P<0.01).Kidney-tonifying Chinese herbs high dose group was better than the other groups(P<0.01).Conclusion:1.Osteoporosis due to kidney-deficiency model can be successfully established by the method of removing the ovaries.2.The expression of PPARγ2 mRNA and protein existed in normal group,which indicate that PPARγ2 existed in bone,kidney and hypothalamus in normal rats,and PPARγ2 may play an important role in bone metabolism.3.In model group,the level of the expression of PPARγ2 mRNA and protein was increased in bone and kidney,the expression of PPARγ2 mRNA and protein was decreased in hypothalamus.These may be one of the important pathomechanisms in osteoporosis.4.Kidney-tonifying Chinese herbs can decrease the level of the expression of PPARγ2 mRNA and protein in bone and kidney,increase the level of PPARγ2 mRNA and protein in hypothalamus,which indicate that kidney-tonifying Chinese herbs can promote visibly inhibit adipose Cell of Kidney deficiency osteoporosis differentiation,it also can promote bone marrow stromal cell differentiate to osteoblast.So we confer that:it might be a new and effective way to increase bone formation,preventing and treating osteoporosis due to kidney deficiency.
