Objective Ischemia cerebral vascular disease is a serious disease which still threatens human health and life quality with higher incidenced disease , higher death and mutilation rate.Now it is generally agreed that apoptosis is the chief death pattern of neurons in penumbra after ischemia and reperfusion caused injury in the brain.To contain apoptosis can reduce the injury of nerve cell in edema band.The acupuncture is predominant in this stage. In this thesis, the protective effects on the brain and the mechanism of acupuncture were studied through the experiments on rats by acupuncture theory and based on the neuron apoptosis and signal transduction. Methods SD rats were randomly divided into normal group, sham operation group,model 24h group, model 48h group , model 72h group, electroacupuncture treatment 24h group, electroacupuncture treatment 48h group, electroacupuncture treatment 72h group. Establish to block the rats middle cerebral artery (MCAO ) -reperfusing model with the suture in MCA."Dazhui","Neiguan" of both side were electroacupunctured. Detected the neuronal apoptosis in hippocampi CA1 by the terminal deoxynucleotidyl transferase mediated dUTP-biotin nickend labeling (TUNEL); observed hippocampus microstructure by HE staining; measured the chang of Tumor necrosis factor-a ( TNF-a ) and interleukin-6 ( IL-6 ) in the hippocampus by radioimmunoassy (RIA); detected nuclearfactouκB ( NF-κB ) and signal transducer and activator of transcription-3 ( STAT3 ), Bcl-2 and Fas distribution in the hippocampi CA1 by immunohistochemistry; measured the chang of NF-kB and STAT3 in the hippocampus by Western blotting, and the effects on above item of electroacupuncture were observed.Results (1)Electroacupuncture may significantly improve the nerve missing sing of the rats with MCAO—reperfiising (P<0.05,P<0.01); release the number of apoptpsis neurons in ischemic hippocampi CA1 (P<0.01,P<0.05 ); improve the pathologic change in neuron of hippocampus and protect the neuron. (2 ) The content of TNF-a and IL-6 in hippocampus of model group rats were significantly higher than those of normal and sham operation group rats (P<0.05 ) ,while after electroacupuncture, the content of TNF-a were significantly decreased with the lapse of time (P<0.05 ); the content of IL-6 were significantly increased (P<0.05 ) .content peak of TNF-a were appeared at 24h,content peak of IL-6 were appeared at 48h. (3 ) The expression of NF-KB-p65 in ischemic hippocampi CA1 and content of NF-κB-p65 in hippocampus of model group rats were significantly higher than those of electroacupuncture group rats (P<0.01,P<0.05) ,and model group rats were significantly higher than those ofnormal and sham operation group rats( P<0.05 ),the expression peak were appeared at 48h,and it were shifted significantly to nucleus. (4) The expression of STAT3 in ischemic hippocampi CAl and content of STAT3 in hippocampus of electroacupuncture group rats were significantly higher than those of model group rats (P<0.01,P<0.05 ) , the expression peak were appeared at 72h,and it were shifted significantly to nucleus; model group rats were significantly higher than those of normal and sham operation group rats (P<0.05 )? ( 5 ) The expression of Bcl-2 in ischemic hippocampi CAl of model group rats were significantly lower than those of electroacupuncture group rats (P<0.01 ) ,and were significantly higher than those of normal and sham operation group rats( P<0.01), the expression peak were appeared at 24h.The expression of Fas in ischemic hippocampi CAl of model group rats were significantly higher than those of electroacupuncture group rats (P<0.01) ,and were significantly higher than those of normal and sham operation group rats( P<0.01), the expression peak.were appeared at 48h.Conclusion NF-κB, STAT3,TNF-α,IL-6,Bcl-2, Fas participate in apoptosis after cerebral ischemis-reperfusion injury.Electroacupuncture adjust the signal transduction of cytokine interposed after cerebral ischemis-reperfusion injury.This effect of electroacupuncture adjusting could be carried...
