ObjectiveNowadays cerebrovascular disease(CVD) has become one of grave diseases to mankind, especially to mid-aged people. With the aging population of the globe more and more increasing, the harm of CVD is increasingly outstanding. It was one of the healthy problem to pay closer attention to preventing and curing acute cerebral accident in the medical public. The attack rate of cerebral arterial thrombosis was far superior to that of hemorrhagic stroke. With the profound investigation into cerebral arterial thrombosis, it could induce the injury of ischemia-reperfusion after bloodstream reviving recanalization. Inflammatory reaction was the cause resulting in cerebral ischemic reperfusion jury. The committed step of Inflammatory reaction was cell adhesion and extravasation of white blood cell -vascular endothelial cell. Intercellular adhesion molecule-1 was important index to review endothelium function. Used the MCAO model, this research investigated the signal transduction of endothelium cell apoptosis mediated by ICAM-1 to research the mechanism of early acupuncture treating resisting cerebral ischemical reperfusion injury after CI. MethodsSD rats were randomly divided into the normal control group , sham operation group, model(ischemia 0.5h reperfusion 24 hours), electroacupuncture treatment(ischemia 0.5h reperfusion 24 hours, treating 2 times). Used the suture in MCA to establish MCAO ischemia-reperfusing model. The remarks of nerve function deficit referring to Lundy remarking standards. The acupoints of BAIHUI and SHUIGOU were selected, stimulated with slow-fast wave for 30 minutes. The rats(some rats from every group)were reperfused with 4% paraformaldehyde after being anesthetized by 10% chloral hydrate, then taken out the brain. The brain tissue 2mm away from the chiasma was cut frontally, the middle part left, immersed in hard paraffin. Observed ischemia brain tissue microstructure, detected the endothelium cell apoptosis in ipsilateral cortex and ipsilateral basal ganglia by the terminal deoxynucleotidyl transferase mediated dUTP-biotin nickend labeling (TUNEL), used immunohistochemistry SABC(spreptavidin-biotin-peroxidase complex) method to investigate the expression of ICAM-1, Bcl-2 and c-fos in rats' ipsilateral cortex and ipsilateral basal ganglia after ischemia-reperfusion and electroacupuncture. In the higher times objects glass, ICAM-1, Bcl-2 and c-fos positive reaction were analysis of accounts. The blood of other rats(others from every group) was collected by extirpating eyealls, Which was put in the room temperation till thrombokinesis, centrifuging 10 minutes, separating serum, conservated in -20 ℃.
