Objective:Proliferation of Tumor cells can be regulated by peripheral blood monounclear cells, cytokines and drugs. Telomeres and telomerase are related to cacinoma cells poliferation. Increased telomerase expression has also been observed in activated lymphocytes. To investigate the role of telomerase of PBMC in the H&NSCCa, we measured telomerase activity during the PBMC proliferation of laryngeal cacinoma patients. Macrophage colony stimulating factor (M-CSF) is a homodimeric growth factor that humorally regulates the growth and differentiation of mononuclear phagocytes. Recent studies have demonstrated overexpression of M-CSF and its receptor in breast, ovarian, and endometrial adenocarcinomas. To investigate the exist of M-CSFR in cell membrane, production of M-CSF and the possible role of M-CSF and its receptor in the laryngeal carcinoma, a prospective study was undertaken to study M-CSF expression in benign and neoplastic laryngeal epithelium and to compare serum M-CSF levels in laryngeal carcinoma patients. Epidemiological investigations, clinical observations and animal studies demonstrate that non-steroidal anti-inflammatory drugs (NSAIDs) can prevent colorectal cancer and breast cancer and squanmous cell cancer. However, the studies of NSAIDs in laryngeal cancer have not been reported. We add aspirin in medium cultured Hep-2 laryngeal carcinoma cells in vitro to elucidate the mechanism of anti-tumor action of aspirin by dection of cell proliferation, apoptosis and cycle.Methods:Thirty-one patients, with histologically confirmed laryngeal carcinoma were included in this study at PUMC Hospital CAMS during Augest 2000 to March 2001.Control samples were obtained from 17 healthy volunteers. Peripheral blood mononuclear cells were separated by Ficoll-Hypaque density gradient. PBMC were activated in vitro with 10 μ g/mL phytohemagglutinin(PHA) for 60 hours. A Commercial telomerase PCR ELISA kit (Roche Molecular Biochemicals Mannheim Germany) was used to assay telomerase activity in all specimens. Cell proliferation was assayed by [3H]-thymidine incorporation. The serum were obtained from 27 laryngeal carcinoma patients. The level of M-CSF.
