Since mid - 70's when indapamide was introduced into the treatment of hypertension, it has received a lot of attention throughout the world. This paper concentrates on the following areas which the past studies have not touched on.1. The pharmacokinetic characteristics of indapamide in the elderly: After taking of indapaiide 1.5 mg, the plasma level of the drug was determinated with HPLC. Compared with the young group) t1/2B is prolonged and Vd, enlarged significantly in the elderly ( both P < 0.05 ). There are no differences of other pharma-cokinetic parameters between two groups. The result shows that the enlargement of Vd accounts for the prolongation of t1/23 in the elderly.2, Ihe hypertensive patients were divided into two groups. One was treated with indapamide ( 2.5 mg / d ) and the other , nifedipine ( 30 mg / d, positive contral ). Two weeks after the drug administration, mean blood pressure in both groups was reduced significantly (P < 0.001 ). In group of indapamide, the serum sodiun and potassium were decreased ( P < 0.05 ) 8 weeks after the treatment, but they were still in the normal ranges; the reduction of blood pressure is accompanied by significant decrease of plasma TXB2 and PGF2a (P < 0.01 and P < 0.001, respectively) and significant increased of plasma 6-Keto-PGF1a and PGE2, and 6-Keto-PGF1a / TCB2 ratio ( P < 0.02, 0.05 and 0.001 respectively ). The result supports the theory that prostaglandins play an important role in the hypotensive...
