There are many kinds of bone substitutes which form new bone by bone conduction. When BMPs are combined with certain carrier, the composite will have activity of bone conduction and bone induction. In this study, the coral and type I collagen were chosen as the carrier of recombinant human bone morphogenetic protein-2 (rhBMP-2). The composite's activity of inducing new bone formation was studied. The maxillofacial bone defects were repaired using the composite of rhBMp-2, coral and collagen. The feasibility of prefabricating certain shape bone autograft was investigated.1. The experimental study on rhBMP-2, coral and collagen composite inducing intramuscle boneIn this study, the composites of rhBMP-2, coral and collagen were implanted in the muscle pouches of mice, rats, rabbits and dogs. The ectopic osteoinductive activity was assessed with history and histomorphometry. The composites induced dose-dependent and time-dependent amounts of intramuscle bone in mice. In rats, the composites induced larger amounts of intramuscle bone than in rabbits and dogs. The collagen may delay the degradation of coral.2. The experimental study on rhBMP-2, coral and type I collagen composite in repairing maxillofacial bone defectsIn this study the composites were applied to repair the rabbits' mandibular defects and implanted at the surface as onlay grafts that was fixed with dental implants. The ability of the composites repairing maxillofacial defects was evaluated with general observation, radiological and histological examination and compared with autogenous bone and coral. In rabbits, new bone was observed earlier and more at the defects after transplantation of the composites and autogenous bone than that of coral, and the reconstruction of the defects with the composites or autogenous bone was superior to that with coral by 2 and 3 months after surgery.
