Two Chinese medicinal herbs, the roots of Sarcococca vagans and the seeds of Allium tuberosum were subjected to phytochemical studies , respectively. 27 compounds were isolated, among them 9 were new constituents . Their structures were established by means of various modern spectroscopic techniques and chemical derivatization. In addition, tandem mass spectrometry (MS/MS) was applied to study the constituents in the total alkaloids from the roots of S. vagans and 10 trace constituents were structurally elucidated by the CED MS/MS of their [M+H]+ ions and 8 of them were new steroidal alkaloids.Phytochemical examination of the roots of S. vagans led to the isolation of seven new steroidal alkaloids , sarcovagine AD (14), sarcovagenine A, B, D (79), together with the known axillarine F (5), pachysamine A (6) and H (10). The structures of the new compounds were determined as 20α-dimethylamino-2β, 4β-dihydroxy-3β-tigloylamino-5α-pregnane (1), 20α-dimethylamino-2α-hydroxy-3β - tigloylamino -4β-acetoxyl -5α-pregnane (2), 20α-dimethylamino-3β-tigloyl amino - 4β - acetoxyl -5α- pregnane(3), 20α-dimethylamino-3-tigloylamino-5α-pregna-2-en-4-one (4), 20a-dimethylamino-2β, 4β-dihydroxy-3β-tigloylamino-5α-pregna-16-ene (7), 20α-dimethylamino-2α-hydroxy-3β-tigloylamino-4β-aetoxyl -5α-pregna-16-ene (8), 20α-dimethylamino-3-tigloylamino-5α-pregna-2,16-dien-4-one (9), respectively. An unusual phenomenon that the carbon and proton signals from C17-side chain were not discernible in the NMR spectra of compounds 15 was discussed.The ESI-MS analysis of the total alkaloids from the roots of S. vagans demonstrated the presence of several trace constituents in the total alkaloids in addition to the isolated compounds.
