The involucres is outermost layer of Castanea mollissima Blume, as a member of Castanea genus of Fagacaea family. It is offen used for antibacterial and anti-inflammatory in civil。The modern experiment also demonstrated that the decoction, EtOH, EtOAC extraction from involucres of Castanea mollissima Blume has antibacterial activity. During the castanea processing, the involucres is always discarded which cause serious waste of the resources. In search of the new biological active substances, we study the chemical constituents from the involucres and the acid hydrolyzed extraction. Based on the idea of semi-bionic extraction, this study on the chemical constituents from involucres of Castanea mollissima Blume acid hydrolyzed.Involucres of Castanea mollissima Blume were extracted wtih the 75% EtOH, then the extraction was processed with acid solution, whose PH is 1.0. The extraction was suspended in water and extracted with petroleum ether,EtoAc and n-BuOH solvent. We obtain three extraction part of the petroleum ether,EtoAc and n-BuOH. By means of the silica gel, sephadex LH-20 column chromatogry isolation methods,14 compounds were obtained. By the methods of NMR and Nature for the structural identification, fourteen compounds were obtained and elucidated as kaempmerol (1), querctin (2),gallic acid(3), protocatechuic acid (4), p-hydroxybenzoic acid(5),5-hydroxymenthyl-furaldehyde (6), emodin (7), kaempferol-3-O-[6",2"-di-O (E)-p-coumaroyl]-β-D-glucopyranoside (8), kaempferol-3-O-[6"-O-(E)-p-coumaroyl]-β-D-glucopyranoside (9), daucosterol (10), isovanillic acid (11),β-sitosterd (12), oleanolic acid (13), ergost-6,22-diene-3β,5a,8a-triol (14) respectively. The compand 11 is the first obtained from the Castanea. We have built an HPLC method to determinate the gallic acid content in the outermost layer of Castanea mollissima Blume. The method is accurate, stable and reliable. We use the mothed to determinate the gallic acid content between hydrolysising before and after. Find that gallic acid content increased significantly after the hydrolysising. We Speculate the increasing gallic acid come from the corresponding tannic acid who are hydrolysised. The extraction method can be as a method which can be used to obtain the gallic acid from the main components of the active site. Our former research discovered that the main chemical substances of involucres form Castanea mollissima Blume are flavonoids and phenolic acid. They have activity of anti-bacterium. Index of gallic acid,by determination of gallic acid in involucres of Castanea mollissima Blume and the acid hydrolyzed by HPLC. The results indicated that the gallic acid level increased obviously by acid hydrolysis. The leaves of Panax ginseng C.A.Meyer is dry leaves of herbal plant belonging to the Aralaceae family. In Chinese Pharmacopoeia, the leaves of ginseng which tastes bitter, cold, sweet have the effects of enriching qi, clearing away heat, activating blood. In order to enlarger the medicinal resource of ginseng, the further study on the chemical constituents of its leaves was carried out. This article studied the extraction of Ginseng leaves in 70% EtOH. Nine compounds were obtained. Identified by spectral analysis, their structures were ginsenoside F1(Ⅰ),20(R)-Rh1(Ⅱ),20(S)-Rhl(Ⅲ),20(S)-protopanaxatriol(IV), 20(R)-Rg3(V),20(S)-Rg2(VI),20(R)-Rg2(Ⅶ),20(R)-Rh2(Ⅷ),20(R)-Rg3(Ⅸ) respectively. The results can provides a reference for the expansion and use of the chestnut, ginseng resources.
