| ObjectiveTo explore the mechanism of the effect of Yifeijiangxian drug on silicosismodel Wista rats by the histopathology of silicosis model Wista rats, the effect ofQi-invigorating and blood-activating, the activity of the SOD and MDA, the contentsof CP and HYP, the observation of the gene expression of TGF- β1 mRNA and typeI type III collagen.Methods1. To all the silicosis model Wista rats, the same amount of physiological saline, Tetrnelrine and Yifeijiangxian drug et al were administered, observing the change of status, body weight, lung weight and lung coefficient of the rats. Then, sacrificed the rats, by the HE staining and Masson staining to rats's lung, to observe the lung histopathologic change under the light microscope.2. By the method of swimming test breath frequency detection and collecting blood from heart, the breath frequency, the time of swimming test, the whole blood viscosity, the PCO2 and PO2 of the model Wista rats were measured.3. Rats were killed with drawing blood from heart. Then make its' refrigerating lung tissues into 10% homogenate, and detect the contents of SOD and MDA in it.4. By the method of colorimetry and alkali hydrolyzing to detect the contents of CP in serum and the contents of HYP in lung.5. By the method of immunohistochemistry and FISH, to detect the gene expression of TGF- β1 mRNA ,type I-collagen, type III-collagen in silicosis model Wista rats lung cell, and then make qualitative and semi-quantitative analysis.Result1.(1) By the HE staining the alveolar inflammation and the pulmonary fibrosis were more mitigated obviously in Yifeijiangxian drug groups than in silicosis model group; after treating for 7 days, pulmonary inflammation cells and infiltration area were more mitigated obviously in Yifeijiangxian drug groups than in silicosis model group; after treating for 14 days, pulmonary inflammation cells were changed lightly and moderately, the pulmonary tissue structure in Yifeijiangxian drug groups was more integrated than in silicosis model group, fibrsblast proliferchion and pathological changes in Yifeijiangxian drug groups was milder than in silicosis model group; after treating for 28 days, the pulmonary fibrosis in Yifeijiangxian drug groups was changed lightly and moderately, the pathological changes were limited, the collagen fibers in pleural membrane and alveolar membrane were bunchy.(2) By the methods of Masson staining that, in Yifeijiangxian drug groups, after treating for 7 days, perivascular and peribronelial collagen proliforation aggradations was discovered; after treating for 14 days, the a few collagen proliforation aggradations were discovered in pulmonary interstitial; after treating for 28 days, the collagen fiber budles were discovered obviously in pulmonary interstitial... |
