Determine The Chromosomal Localization Of The Drosophila Gene Inhaled Anesthetic Sensitivity And Significance

The influence of age of fruit fly on its 50% effective close (ED₅₀) for sevoflurane was observed in this study to determine when is the best age of fruit fly for studing hereditary cross experiments. The different fruit fly strains which were sensitive, normal and resistant to sevoflurane were developed by screening selection so as to offer ideal strains for studies on inhaled anesthetics action. The gene related to inhaled anesthetics sensitivity in above strains was analyzed and determined in chromosome of fruit fly by a serious of crosses to provide information for studying molecular biological mechanisms of inhaled anesthetics action at the level of gene.Study 1:The sevoflurane ED₅₀ of a wild-type Drosophila ( with different day-ages ) was calculated according to a dose-response curve with two methods, one-flask method and brush irritating method, respectively. 480 fruit flies ( with the same day-age ) were placed into four flasks ( each flask contained 120 flies ) and ED₅₀ were measured. An analysis of variance was performed to determine whether there were significant differences among different day-age fruit fly.
