Duanzhuxiaobaqia is the dried rhizome of Heterosmilax yunnanensis Gagnep.which belongs to the genus Heterosmilax Kunth of family Liliaceae. Due to the effects of clearing heat, removing damp, counteracting toxicity, and improving the mobility of joints, it has been widely used for the treatment of Dysuria with turbid discharge, morbid leukorrhea, carbuncle, lymphadentitis, chronic eczema; contracture of limbs and muscle pain in syphilis and leptospirosis, etc. But its chemical constituents have been scarcely reported. In this research, the initial test results showed that Duanzhuxiaobaqia can effectively inhibit the growth of tumor, then systematically studies on its chemical constituents and quality control were carried out, and the investigation for its pharmacological activities, so as to provide theoretical proof and practical reference for its clinical application and further research.1 Studies on the chemical constituents of Duanzhuxiaobaqia: 31 compounds have been isolated from the dried rhizome of Heterosmilax yunnanensis Gagnep. Based on their physicochemical and spectroscopic evidence, 29 structures were elucidated as follows: 8 Flavonoids: daidzein(1), liquiritigenin(2), Hesperetin-7-0-rutimoside(3), Mangiferin(4), 3-O-β-D- glucop yranosyl-1,3,6,7-tetrahydroxy-xanthone(5),7-O-methylmangiferin(6),4-C-β-D-glucopyranosyl-1,3,6-trihydroxy-7-methoxy-xanthone(7), 2-O-α-L- rhamnopyranosyl-5-C-β-D-glucopyranosyl-4,6-dihydroxyl–phenyl -p-hydroxy benzophenone (8); 4 phenolic glycosides: 3,5-dimethoxy-4 -hydroxy-benzoic acid-O-β-D-glucoside (9), 4-hydroxybenzoate glucoside (10), 3,5-dimethoxy-4-hydroxy-1-O-β-D-glucoside(11), glucosyringic acid (12); 4 triterpenoids and sterols: ursolic acid (13),β-sitosterol (14), daucosterol (15),β-sitosteryl-3β-glucopyranoside-6ˊ-O-palmitate (16); 1 anthraquinone: Chrysophanol (17); 1 coumarin: bergapten (18); 8 alcohol,acid and esters :stearic acid (19), Palmitic acid (20), Tetradecanoic acid (21), hexacosanoic acid (22), n- Hextatetranol (23), n- Octacosanol (24), bis(2-ethylhexyl) phthalate(25), glycerol monopalmitate (26);3 other compounds:5-hydroxymethyl furaldehyde (27),n-butyl-β-D-fructopyranoside(28),1-O-[β-D-xylopyranose (1→6)-β-D- glucopyranosyl (1→) methyl acetate (29). Among all these compounds, 5, 8, and 29 are new; except 14,15,19,20and26, it is the first time for the other compounds to be isolated from genus Heterosmilax Kunth; and all the compounds were isolated from this plant for the first time.2 Studies on the quality control of Duanzhuqiaobaqia, including characteristics, micro-construction and TLC identification, quantity determination of total flavonoid by UV-VIS , mangiferin and glucosyringic acid by HPLC have been performed. HPLC fingerprint has been studied too. On this basis, the quality standard of Duanzhuxiaobaqia was drafted.3 Preliminary studies on the biological activities of Duanzhuxiaobaqia. The anti-tumor activities of the n-butyl alcohol extracts have been investigated by in vivo. Moreover, 13 purified compounds obtained from the plants have been assayed on their anti-tumor cell activities by in vitro. The results showed that the n-butyl alcohol extraction of Duanzhuxiaobaqia could significantly inhibit the growth of tumor and parts of purified compounds obtained from the plants could inhibit the activities of tumor cell.In short, in this paper, through the systematical studies mentioned above on Duanzhu xiaobaqia, part of its chemical constituents has been figured out. Its characteristics, micro-construction, TLC, quantitation determination and pharmacology have been studied, as well as its anti-tumor activities. These provided the theoretical basis and scientific reference for further study, development and application of Heterosmilax yunnanensis Gagnep.
