| Dianbaizhu a folk medicine used as an antirheumatic,is made from Gaultheria leucocarpa Blume var.yunnanensis(Franch.)T.Z.Hsu&R.C.Fang(Ericaceae)and has multiple plant origins and officinal parts.Their similar morphologica features lead to difficulty with species discrimination.In addition,there were less researches on quantitative determination of the chemical composition in Dianbaizhu.In most of the researches,only one or a kind of chemical composition was used as an index.Furthermore,chemical constituents remain to be elucidated continuously.The aim of this paper was to found a method to distinguishing between Gaultheria species and ditermined several anti-inflammatory active compounds.Also,the anti-inflammatory extract of Dianbaizhu was conformed and researched by chemical separation method.Firstly,99 reported literatures concerning the researches on G.leucocarpa var.yunnanensis,covering 1977 to 2015,were reviewed.It included the study on plant resources,determination,chemical constituents and pharmacological researches.The objective of this paper was to be a reference for the improvment of Dianbaizhu quality standards.In this study,multiple confusable varieties of Dianbaizhu were distinguished with the ultra high performance liquid chromatograph,data analysis was carried out by the chemometric methods.The methodological study results showed that all RSD values were lower than 2.0%,the classification results were basically consistent with Flora of China.Therefore,this paper provides an idea for the identification of those samples which possessed incomplete morphological characteristics.A variety of inflammatory components existed in G.leucocarpa var.yunnanensis.Here,this study established the method for fast determination of these inflammatory components by using high performance liquid chromatography.The method was precise,stable and reliable.The measurement data was analysed with the chemometrics method.There was a significant difference(P<0.05)between Gaultheria species,and indistinctive differences between medicinal parts(P>0.05).The mainstream species G.leucocarpa var.yunnanensis was the best species(judged by the contant of anti-inflammatory constituents).Overall,the components’ content in branches was higher than that in other three medicinal parts.The results showed that the chromatography technology combined with the chemometrics was very effective for the determination and the data analysis,which provided a reference for the quantitative study on other medicinal herbs.Phytochemical investigation of the inflammatory analgesic active extract(ARF)of G.leucocarpa var.yunnanensis,led to the isolation of 16 known compounds,including four methyl salicylate glycosides Methyl salicylate 2-O-β-D-glucoside(1),Gaultherin(2),MSTG-A(3),MSTG-B(4),three sugar esters compounds Ethyl β-D-xylobioside(5),Ethyl-O-β-D-xylopyranosyl(1-6)-O-β-D-glucopyranoside(6),Methyl-O-β-D-xylopyranosyl(1-6)-O-β-D-glucopyranoside(7),two monoterpene glycosides Roseoside(8),Paeoniflorin(9),five benzoic acids Vanillic acid(10),2,5-dihydroxybenzoic acid(11),3,4-dimethoxycinnamic acid(12),Ferulic acid(13),Chlorogenic acid(14)and two other ingredients 4-hydroxy-2,6-dimethoxyphenol-1-O-β-D-glucopyranoside(15),3-methoxyl-1H-pyrrole(16).Wherein Roseoside(8),Paeoniflorin(9),3,4-dimethoxycinnamic acid(12),4-hydroxy-2,6-dimethoxyphenol-l-O-β-D-glucopyranoside(15)were isolated from the Ericaceae for the first time.Methyl-O-β-D-xylopyranosyl(1-6)-O-β-D-glucopyranoside(7)was isolated from the genus Gaultheria for the first time.The results enriched the type of chemical composition in Ericaceae and also were beneficial to in-depth study of G.leucocarpa var.yunnanensis.This innovative points of this research:1.For the first time,7 confusing species were classificated effectively by using chromatographic techniques with chemometrics methods.2.For the first time,determination research on anti-inflammatory components in Dianbaizhu was carried out by using chromatographic techniques with chemometrics methods.Additionally,It explained the phenomenon,various medicinal parts used to treat rheumatism by many ethnic minorities.In addition,the results proved that 30%ethanol extract was anti-inflammatory and analgesic active part(ARF)and the anti-inflammatory effect come from several anti-inflammatory compounds. |
