Objective To explore the effects and the mechanisms of Astragulas Membranaceus in rats with renovascular hypertension.Methods 30 male SD rats weighing 200-250g were divided randomly into three groups:Astragulas Membranaceus group,control group, sham-opration group.There are were 10 rats in each group.Except sham-opration group,the other two groups were made with two kidney,one clip(2K1C)opration.Blood pressure was measured before opration;After operation,blood pressure was measured once each 4 weeks.4 weeks After opration,Astragulas Membranaceus group were treated with Astragulas Membranaceus(8g.kg-l.d-1,ip),the other 2 groups were treated with an equal volume salt solution(0.9%).8 weeks after opration,the animals were killed.Left ventricular mass index(LVMI),left ventricular wall thickness(LVWT), interventricular septal thickness(IVST),left ventricular diameter(LVD), cardiomyocytes diameter,collagen volume fraction(CVF),perivascular volume collagen area(PVCA)were measured.The protein expression of extra-cellular signal regulated kinase 1(ERK1)were detected by immunohistochemical method.The protein expression of protein-rich tyrosine kinase 2(PYK2)and protein kinase B(PKB)were detected by westem-blot.The protein expression of intercellular adhesion molecule-1(ICAM-1)were detected by ELISA.Results(1)4 weeks after opration,blood pressure in control group and Astragulas Membranaceus group were more than 150 mmhg.4 weeks after opration,compared with sham-opration group,blood pressure in control group were significantly higher(P<0.05);Compared with control group,blood pressure in Astragulas Membranaceus group were no difference(P>0.05)(2)Compared LVMI,LVWT,IVST,LVD in three groups:LVMI(mg/g)in sham-opration group,control group,Astragulas Membranaceus group were 2.7±0.24,3.42±0.26,3.13±0.23;LVWT(mm)in sham-opration group,control group,Astragulas Membranaceus group were 2.25±0.42,4.26±0.48,3.28±0.36;IVST(mm)in sham-opration group,control group,Astragulas Membranaceus group were 2.13±0.38,3.98±0.32,3.02±0.28;LVD(mm)in sham-opration group,control group,Astragulas Membranaceus group were 3.76±0.29,2.18±0.27,2.82±0.20.LVMI,LVWT,IVST in control group were significantly higher than those in sham-opration(P<0.05)LVD in control group were significantlY lower than that in sham-opration group(P<0.05) LVMI,LVWT,IVST in Astragulas Membranaceus were significantly lower than those in control group(P<0.05).LVD in Astragulas Membranaceus group significantly higher than that in control group(P<0.05)(3)Compared cardiomyocytes diameter,CVF,PVCA in three groups: Cardiomyocytes diameter(μm)in sham-opration group,control group, Astragulas Membranaceus group were 14.54±2.25,19.56±2.53,16.58±2.46; CVF(%)in sham-opration group,control group,Astragulas Membranaceus group were 3.83±1.40,11.21±2.96,7.83±1.67;PVCA(%)in sham-opration group,control group,Astragulas Membranaceus group were 15.71±3.85,30.58±6.25,21.764±4.36.Cardiomyocytes diameter,CVF,PVCA in control group were significantly higher than those in sham-opration group(P<0.05) cardiomyocytes diameter,CVF,PVCA in Astragulas Membranaceus group were significantly lower than those in control group(P<0.05).(4)Compared protein expressin of ERK1 in three groups:protein expressin of ERK1 in sham-opration group,control group,Astragulas Membranaceus group were 0.15±0.02,0.20±0.04,0.16±0.03.Protein expressin of ERK1 in control group were significantly higher than that in sham-opration group (P<0.05).Protein expressin of ERK1 in Astragulas Membranaceus group were significantly lower that that in control group(P<0.05)(5)Compared protein expression of PYK2 and PKB in three groups: Protein expressin of PYK2 in sham-opration group,control group,Astragulas Membranaceus group were 0.47±0.08,0.76±0.12,0.61±0.09;Protein expressin of PKB in sham-opration group,control group,Astragulas Membranaceus group were 0.52±0.07,0.79±0.11,0.67±0.08.Protein expressin of PYK2 and PKB in control group were signifcantly higher than those in sham-opration group(P<0.05).Protein expressin of PYK2 and PKB in Astragulas Membranaceus group were significantly lower than those in control group(P<0.05).(6)Compared protein expression of ICAM-1 in three groups:Protein expression of ICAM-1 in sham-opration group,control group,Astragulas Membranaceus group were 0.13±0.02,0.21±0.05,0.16±0.03.Protein expression of ICAM-1 in control group were significantly higher than that in sham-opration group(P<0.05).Protein expression of ICAM-1 inAstragulas Membranaceus group were significantly lower than that in control group (P<0.05)Conclusions(1)In this study,the model of left ventricular hypertrophy in rats with renavascular hypertension using 2K1C opration was stable and successful.(2)LVH in renavascular hypertension might be caused by the increased expression of ERK 1,PYK2,PKB protein.(3)LVH in renalvascular hypertension might be associated with the increased expression of ICAM-1 protein and the imflammatory reaction mediated by ICAM-1.(4)The treatment of Astragulas Membranaceus can reverse LVH and the myocardial interstitial fibrosis.(5)The treatment of Astragulas Membranaceus can reverse LVH and the myocardial interstitial fibrosis,which might through the decreased expression of ERK1,PYK2,PKB protein and inhibition of imflammatory reaction mediated by ICAM-1.
