(1)Objective In order to study effect of boxintong capsule on people with angina and the relating serum indexes。Methods Forty patients were divided randomly into two groups:the first group which contained 20 people took with Boxintong capsule;another 20 people took with Suxiaojiuxin pill.The period of treatment was four weeks.Results after the treatment,the validity of Boxintong group was close to the su xiao jiu xin wan group,the level of the platelet maximum agglutination fell the other group(p<0.05);the level of SOD and NO increased(p<0.01);the level of IL-6 in Boxintong group fell surpass the other group(P<0.05);the level of IL-1,MDA,TNF-αand CRP fell(p<0.05).Conclusions Boxintong capsule could depress platelet maximum agglutination and decrease angina symptoms.The mechanism is possibly about accelerating the synthesize of NO and SOD,depressing the level of the MDA,distending the coronary artery,resisting inflammation,Lowering the low density lipoprotein cholesterol.(2)Objective To illuminate the mechanism of Boxintong capsule on treating acute myocardial ischemic rabbits by observing the influence on electrocardiogram in acute myocardial ischemic rabbits.Methods Taking the acute myocardial ischemic rabbits as experimental object,it is compared with suxiao jiuxinwan group and control group,observing the influence of Boxintong capsule on the number of Notching,variety of heart rate,transform of ST-T segment,change of arrhythmia wrong and heart rate variability in acute myocardial ischemic rabbits.Results All the inspective targets of acute myocardial ischemic in rabbits have evident amelioration compared with the Suxiao jiuxinwan(P<0.05). Conclusions Boxintong capsule is usable in treating the acute myocardial ischemic rabbits,but the molecular mechanism need to study more.(3)Objective To observe the effect of the serum containing Hua-xia shallot on low glucose and hypoxia injury on the rat cardiomyocytes.Methods Cardiomyocytes of rats were separated in vitro and cultured with pancreatin and preplating technique,then randomly divided into 7groups:Control group,low glucose and hypoxia group,The different density(40%,30%,20%,10%)serum of Hua-xia shallot group;50μg/mlEnalapril group.We assay myocardial viability by MTT techique,detect the level of SOD and MDA.Results The serum of Hua-xia shallot group all can increase SOD activity remarkably and inhabite MDA production significantlyduring hypoxia injury.The SOD level of the serum containing 30%Hua-xia shallot is close to normal,serum group,better than enalapril(P<0.05).Conclusion The serum containing 30%Hua-xia shallot can improve the role of oxygen-glucose deprivation myocardial cells antioxidant enzyme synthesis,decrease myocardial cell damage caused by oxygen-glucose deprivation.(4)Objective To study the effects and mechanism of Hua-xia shallot on combating atherosclerosis.Methods 32 New Zealand white rabbits were randomly divided into 4 groups:①normal group②Model group③For medicine group④suxiao jiuxinwan group.Detection of apoptosis-related genes and biochemical indicators such as NO et al.Results Hua-xia shallot can adjust electrocardiogram better than suxiao jiuxinwan.Hua-xia shallot can increase the xepression of NO,6-keto-PGF1α,SOD,bcl-2(P<0.01,P<0.05),decrease the xepression ofET-1,TXB2,MDA,CRP,OX-LDL,TC,TG,AI index,PDGF-B,bFGF,TGF-β1,VEGF,fas,c-myc(P<0.01,P<0.05).Hua-xia shallot can lower IMT and control the apoptosis of VSMC bettr than suxiao jiuxinwan.Conclusion Hua-xia shallot can combat atherosclerosis.(5)Objective To observe the influence of Hua-xia shallot on restenosis after interventional therapy.Methods 80 Wistar rats were randomly divided into 8 groups:model group(2w/4w),Hua-xia shallot group(2w/4w),valsartan group(2w /4w),Lipitor group(2w/4w),10 rats in each group.Rats were treated with drugs and high-fat foods for totally 2 or 4 weeks.Immunohistochemistry method was used to detect the expression of matric metallo protenase-2(MMP-2),p roliferation cell nuclear antigen(PCNA)and typeⅠcollagen(CPⅠ)in the vascular.The expression were analyzed by imane analysis.Results At each time point,the lumem area was larger and the neointima area was smaller in all the treated groups than that in model groups(P<0.05).The expression of MMP-2,PCNA and CPⅠin the vascular in all the treated groups was decreased,that of the Hua-xia shallot group(2w,4w) being the lowest(P<0.05).Conclusion Hua-xia shallot has the effect of preventing restenosis after balloon injury.(6)Objective To observe the effect of Hua-xia shallot preparation on human umbilical artery smooth muscle cells proliferation induced by AngiotensinⅡin culture and to find the possible molecular mechanism.Methods Culturing the human umbilical artery smooth muscle cells;the number and activity of cells were observed by micrography and MTT technique;the SOD and NO contents were assayed by respective kit;the protein expression and their mRNA levels of PPAR-γand NF-κB were measured by western blot technique and reverse transcription polymerase chain reaction(RT-PCR).Results Comparison to the control group, AngiotensinⅡdecreased the apoptosis level of cells and propeled the cells proliferation.These effects induced by AngiotensinⅡwere all significantly inhibited by Hua-xia shallot preparation.It can up-regulated the protein and mRNA levels of PPAR-γand NF-κB dramatically.Hua-xia shallot preparation can increase the apoptosis level of cells and inhibited the cells' proliferation.Conclusions AngiotensinⅡmay be involved in the progression of atherosclerosis by causing the smooth muscle cells proliferation directly.Hua-xia shallot preparation can protect against AngiotensinⅡinducing smooth muscle cells proliferation by up-regulating the protein and mRNA levels of PPAR-γand down-regulating the protein and mRNA levels of NF-κB,which suggests that Hua-xia shallot preparation may play a role in the prevention and treatment cardiovascular disease.(7)Objective To observe the protective effect of Hua-xia shallot preparation on human umbilical vein endothelial cell injury induced by ox-LDL in culture and to find the possible molecular mechanism.Methods Preparation and identification of ox-LDL and culturing the human umbilical vein endothelial cell;the number and activity of cells were observed by micrography and MTT technique;the SOD and NO contents were assayed by respective kit;the protein expression and their mRNA levels of PPAR-γand eNOS were measured by western blot technique and reverse transcription polymerase chain reaction(RT-PCR).Results Comparison to the control group,ox-LDL increased the apoptosis level of cells and decreased the nitric oxide releases.These effects induced by ox-LDL were all significantly inhibited by Hua-xia shallot preparation.It can up-regulated the protein and mRNA levels of PPAR-γand eNOS dramatically.Hua-xia shallot preparation can decrease the apoptosis level of cells.Conclusions ox-LDL may be involved in the initiation and progression of atherosclerosis by injuring the endothelial cells directly and causing endothelial dysfunction,Hua-xia shallot preparation can protect against ox-LDL inducing endothelial cell injury by decreasing the protein and mRNA levels of PPAR-γand eNOS,which suggests that Hua-xia shallot preparation may play a role in preventing and treating cardiovascular disease.(8)Objective To study the effects of Hua-xia shallot on the myocardial viability after myocardial infarction in the pigs.Methods 18 myocardial infarction pigs were randomly divided into 3 groups:①Model group(n=6)②Hua-xia shallot group(n=6)③Trimetazidine group(n=6),besides,we take 2 normal pigs as control group.We detected myocardial infarction area,myocardial apoptosis,myocardial cell metabolism,heart wall motion.Results From the electrocardiogram we saw that ST-segment drop to equipotential lines,pathological Q-wave disappeare after 20 min treated with Hua-xia shallot.15 minutes later,cTnI began to increase after plugging LAD.Four weeks later,3 piges format aneurysm,Kinesis significantly weakened in 3 pigs,Kinesis sligtly weakened in the other 12 pigs.Conclusion Hua-xia shallot prepration can correct ischemic state in the Pigs' acute myocardial infarction districts,Save myocardial viability after myocardial infarction.
