BackgroudDF have had over 200 years at least and many times pandemics., The popular scale and frequency of DF have expanded endlessly since 20 centuries. Eevery year there are two thousand five hundred million people suffer the threaten from DF. Urbanization's expand, population's growth quickly, inadequate garbage disposal, augmented international tour, vaccine's deficiency , indistinct prevalence regularity and pathogenesy . absent specific treatment, all these make DF's popular hard to control. The incidence rate of central nervous system (C.N.S) pathological changes for DF sick is 1.6-4.3 percent. The incidence rate of. Dengue encephalitis(DE) is 3~5 times for those people that have already C.N.S pathological changes, and the patients with DE is much easier to happen DHF/DSS.DF vaccinoprophylaxis is still in the trial, and in the near future there are no vaccines yet for being used generally.There are no special effective method for DF's treatment at present, thus to study its symptom-cure feature is very important at the earlier period of DF with TCM 's preponderance to resist viral disease, with the combination of differential diagnosis of diseases and differentiation of symptoms and signs.ObjectiveOn the base of constructing BALB/C mice model of damp-heat syndrome for DV infection, to study dengue fever damp-heat feature and the mechanism of clearing heat and promoting urination method to resist the damage of blood-brain barrier for dengue encephalitis, and to guide the prevention and cure for dengue fever and dengue encephalitis.Methods1 Clinical researchTo collect 1200 dengue fever cases from Canton city NO.8 people hospital according to the sequence from near to distant, and to renew authorizing after verifying according to the sequence of notes-combing(to unify differentiation of symptoms and signs )-building library-table rank-statistics. For climate characteristic, clinical manifestation, examination result, complication and syndrome of 1200 dengue fever cases, adopting SPSSV13.0 statistical package to undertake statistical analysis.2 Empirical studyAt first, to grope the condition through four experiments—high grease forage feeding four days to form internal damp-heat; Temperature 32±0.5℃, humidity 60±5%, and to interfere in two hours to form exogenous damp-heat; Intracal. Inoculation for DV, and virus dilute strength 10-3; At the same time of subculturing DV, to feed high grease forage, then to plus exogenous damp-heat at the 5th days, or first feed with high grease forage, and then simultaneously adding to DV infection and high grease forage feeding.Then to construct the BALB/C mice exogenous-endogenous damp-heat syndrome model for dengue viral infection with high grease forage , environment and dengue viral Intracal inoculation.56BALB/C mice are divided into 7 groups, 8 per. Domesticating 2 days according to convention before intervention.A normal groupB DV infection groupC DV infection with exogenous damp-heatD DV infection with endogenous damp-heatE DV infection with exogenous-endogenous damp-heatF The traditional Chinese medicine group 1 (abrotani herba and baical skullcap root cholagogue soup to interfere in)G The traditional Chinese medicine group 2(Lonicerae and Forsythiae Powder to remove arctii,fructus, kakuda figwort root, and to add apricot seed, French chalinoplasty recipe to interfere in)Adopting the means which at the same time of subculturing DV, to feed with high grease forage, then to plus exogenous damp-heat at the 5th days.Except Group A, At the 1st day, Group B, C, D, E, F and G mice are all inoculated with DV by Intracal, and Group D, E, F and G mice are feeded 4 days with high grease forage simultaneously. Group C, E, F and G mice are put into artificial climate room every other day(the 5th ,7th, and 9th days), and group F and G mice are interfered with the traditional Chinese medicine. The 9th day, to dissect all mice and detect corresponding indicatrix.The detection of body weight and rectal temperature are carried out at the 1st ,5th ,7th , and 9th days respectively. Adopting SPSS13.0 software statistics parcel to analyze experimental results.Results1 Clinical findingsThe patient's condition is light totally forl200 dengue fever cases, and severe cases are few. Brain fever happen after course '4~6days .clinical manifestation: severe headache, delire, neck rigidity, Kernig or Brudzinski masculine. After 2~4 days treatment, symptom is relieved obviously, no sequel.The differentiation of symptoms and signs of TCM is mainly damp-heat, and the damp-heat pathogenic factor is the main causative agent. The pattern of syndrome is mostly damp-heat syndrome.2 Experiment resultsDV infection group(Group B) shows up activity and foodintake to reduce temporarily, body weight no marked change, rectal temperature going up and going down suddenly. The average optical express level of NF-κB in liver and brain tissue , and AQP4, MMP9 and GLUT1 in brain tissue go up. The tissue slices for liver and brain show hepatic and brain cell damage slightly.DV infection with exogenous damp-heat(Group C) and DV infection with endogenous damp-heat(Group D) show up rectal temperature going up and down suddenly. The average optical express level of NF-κB in liver and brain tissue , and AQP4, MMP9 and GLUT1 in brain tissue go up. The tissue slices for liver and brain show the midrange damage of hepatic cell and the slight damage of brain cell.DV infection with exogenous-endogenous damp-heat (E group) show up the average optical express level of NF-κB in liver and brain tissue , and AQP4, MMP9 and GLUT1 in brain tissue to go up obviously. The tissue slices for liver and brain show hepatic and brain cell damage obviously.The traditional Chinese medicine group 1 (abrotani herba and baical skullcap root cholagogue soup to interfere in) and the traditional Chinese medicine group 2 (Lonicerae and Forsythiae Powder to remove arctii,fructus, kakuda figwort root, and to add apricot seed, French chalinoplasty recipe to interfere in) show up the average optical express level of NF-kB in liver and brain tissue , and AQP4, MMP9 and GLUT1 in brain tissue no marked change. The tissue slices for liver and brain show hepatic and brain cell normal roughly.Conclusion1 DF' development and popular have a close touch with environmental factor. Those who have damp-heat constitution are susceptible population and easier to take place dengue encephalitis in humid heat surroundings. The pattern of syndrome for dengue encephalitis is damp-heat fumigating and DV trapping into the upper orifices, and it is a pilot process from start to DHF/DSS.2 The appearance of simple DV infection mice is similar to mitis DF, DV infection with exogenous damp-heat group mice or DV infection with endogenous damp-heat group mice to typical DF, and DV infection with exogenous-endogenous damp-heat to gravis type DF.3 With the pristine intervention of abrotani herba and baical skullcap root cholagogue soup and Lonicerae and Forsythiae Powder to remove arctii,fructus, kakuda figwort root, and to add apricot seed, French chalinoplasty recipe, the average optical express level of NF-κB , AQP4, MMP9 and GLUT1 don't go up, BBB is exempted to be destroyed, and the development of dengue encephalitis is prevented.
