Objective:By means of the observation of the expression of platelet membrane glycoprotein(GP)FIB-R,CD62P and asymmetric dimethylarginine(ADMA)in patients with acute ischemic infarction,the influence of platelet GP FIB-R,CD62P and ADMA on acute cerebral infarction,and the correlation among them were studied, the possibility of using GP FIB-R,CD62P as means to monitor the activated degree of platelet and to provide indirect evidence for pathogenetic condition were explored.These findings were expected to lead to a new strategy of antiplatelet therapy for acute cerebral infarction.Methods:1.The expression of GP FIB-R,CD62P were studied by flow cytometry(FCM)to observe their changes in patients with acute cerebral infarction.2. High performance liquid chromatography(HPLC)was used to detect the level of plasma ADMA in patients with acute cerebral infarction.3.Linear correlation coefficient was calculated to examine the correlation among the expression of GP FIB-R,CD62P and vascular risk factors and prognostic factors.Multiple logistic regression was used to estimate the influence of ADMA on acute cerebral infarction.Result:1.GP FIB-R,CD62P expression were increased in patients acute cerebral infarction more than that in control group.2.A high rate of increased platelet GP FIB-R,CD62P expression was found in stroke patients complicated with hypertension(HP),coronary artery disease(CAD),diabetes mellitus(DM),it indicated these risk factors may promote GP FIB-R expression,increaseplatelet activated degree and induce occurrence of cerebral infarction.But this study didn't show differences significantly in FIB-R,CD62 expression among stroke patients with HP,CAD, and DM with each other.3.FIB-R,CD62P showed significant line positive correlation with neurologic impairment score in patients with acute ischemic stroke,it indicated that platelet activated degree can reflect pathogenetic condition.4.FIB-R,CD62P expression increased in progressive cerebral infarction patients more than stable cerebral infarction,it indicated that FIB-R,CD62P can be used as the indexes for occurrence,development and pathogenetic condition changes of acute cerebral infarction.5.FIB-R,CD62P expression was significant line positive correlation with size and location of cerebral infarction.6.The result that FIB-R,CD62P expression showed significant positive correlation with Rankin scale,indicated that platelet activated degree was close correlation with prognosis of cerebral infarction.7.FIB-R,CD62P expression was close correlation with platelet parameter.8.Plasma ADMA concentration and platelet GP FIB-R,CD62P expression increased in acute cerebral infarction patients more than control group.9.Traditional risk factor including HP,DM,cholesterol was close correlation with cerebral infarction.Plasma ADMA concentration was correlation with acute cerebral infarction.10.Plasma ADMA concentration Was positive correlation with age,hypertension and GP FIB-R,CD62P expression,this findings indicated that GP FIB-R,CD62P increased when Plasma ADMA concentration increased.Conclusion:The study found that platelet glucoprotein FIB-R,CD62P by FCM can be used as sensitive indexes of platelet activated degree.Platelet activation plays an important role in the process of acute cerebral infarction.FIB-R,CD62P expression was positive correlation with progression of acute cerebral infarction. They can influence the prognosis of cerebral infarction.MPV PDW of Platelet can be also used as indexes of platelet activation.Plasma ADMA concentration and FIB-R, CD62P also play certain role in the occurrence and development of acute cerebral infarction.Plasma ADMA concentration can be one of risk factors of cerebral vascular disease.Plasma ADMA concentration was positive correlation with FIB-R,CD62P, These evidences support that causal relevance between plasma ADMA concentration and cerebral infarction exist by its influence on platelet activation.
