Objective: To investigate the effect of Gongqing Granula combined with Mifepristone on the human chorionic villi and decidual cells'apoptosis, as well as the expression of apoptotic gene in early pregnancy .To explore the mechanism of this prescription in preventing and treating bleeding after medical abortion.Methods:180 early pregnancy women who wanted to terminate voluntarily their pregnancy were divided randomly into 3 groups: the test group (Gongqing granula combined with Mifepristone),the control group( Qianzhi capsule combined with Mifepristone), the empty group(Mifepristone) ,compared to artificial abortion group. The clinical effect of medical abortion were observed and chorionic villis and decidual tissues were collected. Transmission microscope was used to give the chorionic villi and decidual tissue a morphology qualitative observation and Annexin V staining joint PI for the quantitative detection of apoptosis. The expression of apoptosis genes(bcl-2/bax,wtp53,c-myc) in human chorionic villi and decidual were detected in the meantime by immunohisto chemical staining(SP method )and in situ hybridization.Results: The clinical experimental investigation showed that the clinical curative effect of Gongqing granula was obviously better than other 2 groups(P<0.05). The observation of ultrastructure indicated distinct change of apoptosis.Compared to the other 3 groups,the expression of bax,baxmRNA,wtp53 and c-myc of the test group were higher than other 3 groups(P<0.05),while the expression of bcl-2 and bcl-2mRNA of the test group was lower than other 3 groups(P<0.05).Conclusions:Gongqing Granula can raise the rate of human chorionic villi and decidual cells'apoptosis to reduce the rate of bleeding after medical abortion and improve the success rate of medical abortion by enhancing the expression of the medical abortion'chorionic villi and decidual pro-apoptotic genes(bax,baxmRNA,wtp53 and c-myc)and decreasing the expression of bcl-2 and bcl-2mRNA.
