| Baekgroud:Former experiments have identified some differentially expressed proteins between MF patients and healthy controls by SELDI-TOF-MS,2-DE and MALDI-TOF-MS.3 of them appear in multigroup of MALDI-TOF-MS:S100A8 which accords with SELDI-TOF-MS,SOD2 and FABP5.Purpose:To testify the differential expression of the 3 proteins above in MF tissues and healthy controls.Method:Extract the protein of eyelids in controls and MF tissues in patients,then confirm the differential expression with Western blot.Do IP simultaneously with MF tissue lysates,and use the supernatants pre-and-after IP to identify the peak at 10837m/z is S100A8 with SELDI-TOF-MS.Finally,localize the expression of the 3 proteins in tissues with IHC and IF.Result:S100A8,SOD2 and FABP5 are overexpressed in MF lesional tissues.Conelusion:We have verified the result of mass spectrum,and layed the foundation of searching for tumor markers. |
