In this doctoral dissertation,the phytochemical and pharmacological studies were carried out on the medicinal plants of Sarcandra glabra.Meanwhile,the advance in the recent research of chemical constituents and pharmacological activities of Sarcandra glabra were reviewed.In our research,the whole plant of Sarcandra glabra was studied systemically in chemistry by means of chromatography methods and spectral technologies.Fifty-six compounds were isolated from the 95%EtOH extract and acetone extract.Fity compounds were determined by spectroscopic analysis and chemical methods.As a result, there were 16 sesquiterpenoids,10 phenolic acids,8 coumarins,5 flavonoids and 3 lignans,one and seven other compounds.Their chemical structures were elucidated as follows:16 sesquiterpenoids:sarcaglaboside F(CSH01),sarcaglaboside G(CSH02), sarcaglaboside H(CSH03),chloranoside A(CSH04),sarcaglaboside C(CSH05) sarcaglaboside D(CSH06),atractylenolideⅣ(CSH07),chloranthalactone B(CSH08), chloranthalactone E(CSH09),9-hydroxy-heterogorgiolide(CSH10),8β,9α-dihydroxy lindan-4(5),7(11)-dien-8α,12-olide(CSH11),shizukanolide E(CSH12),shizukanolide F (CSH13),dihydrovomifoliol-O-β-D-glucopyranoside(CSH14),drovomifoliol-O-β-D-glucopyranoside (CSH15),dihydrovomifoliol(CSH16);8 coumarins:6,7-dimethoxy-coumarin -8-ethleneglyol-2'-O-β-D-glucopyranoside(CSH17),fraxin(CSH18),scopolin (CSH19),eleuthe roside B1(CSH20),fraxidin-8-O-β-D-glucoside(CSH21),isofraxidin (CSH22),fraxidin(CSH23),esculetin(CSH24); 3 lignans:(-)-(7S,8R)-dihydrodehydrodiconiferyalcohol(CSH25),(-)-(7S,8R)-dihydro dehydrodiconiferyalcohol 4-O-β-D-glucopyranoside(CSH26),(-)-(7S,8R)-dihydrode hydrodiconiferyalcohol 9'-O-β-D-glucopyranoside(CSH27);5 flavonoids:naringenin -6-C-β-D-glucopyranoside(CSH28),naringenin-8-C-β-D-glucopyranoside(CSH29), 5,7,3',4'-quadrihydroxy-dihyflavanones-3-rhamnoside(CSH30),5,7,3',5'-quadrihydroxy -dihyflavanones-3-rhamnoside(CSH31),eucryphin(CSH32);11 phenolic acids: evofolin A-8-O-β-D-glucopyranoside(CSH33),β-hydroxypropiovanillone(CSH34), 3,5-dimethoxy-4-hydroxy phenyl)methyl-O-β-D-glucopyranoside(CSH35),glucosyringic acid(CSH36),caffeic acid(CSH37),isovanillic acid(CSH38),protocatechuic acid (CSH39),quinic acid(CSH40),shikimic acid(CSH41), 4,5,6-trihydroxy-methylformatecyclohexene(CSH42),methyl rosmarinic(CSH43),;7 other compounds:sarcaglabetone A(CSH44),sarcaglabdiol A(CSH45),hentriacontane (CSH46),palmitic acid(CSH47),β-sitosterol(CSH48),β-galactose(CSH49),sourose (CSH50).Among these compounds,8 of which were new compounds:4 sesquiterpenoids, CSH01,CSH02,CSH03,CSH07;1 coumarine:CSH17;2 phenolic acids:CSH33, CSH34 and other compound CSH44,CSH45.Compounds CSH10,CSH12,CSH13, CSH14,CSH15,CSH16,CSH18,CSH19,CSH35,CSH36,CSH40,CSH41,CSH42, CSH46,CSH49 were isolated from this genus for the first time.The cytotoxicity of some compounds was studied with MTT methods.Compounds CSH10,CSH08和CSH20 showed strong activity.Compounds CSH03,CSH06 and CSH04 were studied on the antitumour and antiinfective bioactivities by computer aids drug screening and estimated to have antiinfective bioactivity.The target was predicted to be candida albicans dihydrofolate reductase,bacterial dihydrofolate reductase and serratia marcescens chitinase.Compound CSH06 was estimated to have oncolytic activity and had the capaciblity of combining with cytochrome P450 2C9.The extraction and separation method for sesquiterpenes from Sarcandra glabra was summarized.The spectroscopic feature of linderene and eduesmane sesquiterpenes was summarized.
